    Transcriptome sequencing-based study of gene expression differences in resistance to Aeromonas hydrophila infection in Megalobrama terminalis and Megalobrama amblycephala

    To investigate the differences in resistance to Aeromonas hydrophila infection between Megalobrama terminalis and M.amblycephala,the transcriptome of the infected M.terminalis and M.amblycephala was sequenced by the Illumina HiSeq sequencing platform.The results showed that 62 780 Unigenes were obtained from the samples of M.terminalis and M.amblycephala after sequencing and filtering,with an average length of 531.883 bp,and the N50 of Unigenes was 652 bp.The NR and COG database identified 35 443 and 8 632 Unigenes,respectively,accounting for 56.46 and 13.75 percent of all annotated genes.Among the 62 780 Unigenes,33 693 Unigenes were expressed in M.terminalis and 32 936 Unigenes were expressed in M.amblycephala,of which 24 834 Unigenes were co-expressed in M.terminalis and M.amblycephala.Cluster analysis of differential gene expression trends revealed that the gene expression trends of M.terminalis and M.amblycephala at 3 h and 24 h could be divided into 6 categories,with cluster mode 4 containing the most differentially expressed genes(16),cluster mode 1 containing the second-most(15),and cluster mode 5 containing the fewest(4).GO enrichment analysis showed that the differentially expressed genes of M.terminalis and M.amblycephala after infection mainly were concentrated on GO items such as immune response(GO:0006955),antigen processing and presentation(GO:0019882),and membrane(GO:0016020).Enrichment analysis of the KEGG pathway showed that Autoimmune thyroid disease(ko05320),Type I diabetes mellitus(ko04940),Viral myocarditis(ko05416),Cell adhesion molecules(ko04514),and Antigen processing and presentation(ko04612) pathways were highly enriched.The GO and KEGG enrichment analysis showed that the gene expression difference between M.terminalis and M.amblycephala after infection was closely related to antigen presentation genes.This study provided a basis for analyzing the differences between M.terminalis and M.amblycephala immune response during the A.hydrophila infection.

    Macrobenthos are benthic ecological groups that live on the bottom or surface of Marine sediments.As an important part of marine ecosystem,macrobenthos usually have weak locomotion ability and are widely used to indicate the quality of their dwelling environment.It is of great value to study the community structure characteristics and diversity index of macrobenthos for the protection of marine environment and biodiversity resources.In order to study the community characteristic of macrobenthos in coastal area of southern Fujian Province,the investigation and sampling of macrobenthos were conducted in summer 2018.The species composition,density,biomass,biodiversity,community structure and Abundance/Biomass Curves of the macrobenthos were calculated and analyzed.The results showed that 126 species in 11 phyla were collected in the survey,most of them were annelids,followed by arachnids and mollusks.The dominant species were Prionospio malmgreni, Scalibregma inflatum and Aglaophamus dibranchis.The density and biomass distribution were very uneven,the mean values were(374±322) ind/m2 and(12.97±23.16) g/m2,respectively.Evenness index,richness and diversity index were at a high level.There was a significant negative correlation between the number of species at each station and pH,salinity,and a significant positive correlation between the number of species at each station and total nitrogen.The richness index was negatively correlated with pH and salinity,and positively correlated with total nitrogen.There was a significant negative correlation between density and salinity.There was a significant negative correlation between biomass and pH,salinity,and an extremely significant positive correlation between biomass and chlorophyll-a.Macrobenthic communities could be divided into four groups.In this study,there was a lack of characteristic species with large body size,and polychaetes with small body size were dominant spieces.ABC analysis showed that the community structure of macrobenthos was stable.Compared with historical data,the density,diversity and richness of macrobenthos in the coastal area of southern Fujian were higher than those in the northern Fujian,and the biomass and evenness were in the same level.The community structure of macrobenthos in the coastal area of southern Fujian was also more complex and the biodiversity was higher.

    The study of water environment pollution is an important means to control watershed pollution.Taking the Minjiang River basin as an example,the trend of water quality change at 16 monitoring stations in the study area from 2004 to 2020 was analyzed by using the Seasonal Kendall Trend Analysis method,and the main pollution identification and analysis of 13 monitoring points from 2017 to 2020 were conducted by using the Time Series Global Principal Component Analysis method.The results showed that the pH and DO of tributaries were higher than those of the main stream,the main stream was lower in the upper and middle reaches and higher in the lower reaches,COD and NH3-N decreased first and then increased in the main stream from the upper reaches to the lower reaches,TP increased first and then decreased.In addition,Kendall’s test showed an improving trend in pH and DO in the study area,while TP and NH3-N were at higher risk of contamination and some sites needed more attention to COD.Finally,Time Series Global Principal Component Analysis was used to identify that the current water pollution pressure in the study area mainly comes from NH3-N and TP,especially the pollution control of tributaries in the basin needed to be strengthened.

    Comprehensive evaluation of environmental quality in Hainan Lingshui Xincun Port mariculture area

    Based on the data of water quality,sediment,and biological indicators of Xincun Port’s mariculture area from 2011 to 2018,the single factor pollution index,comprehensive environmental quality index and eutrophication index were used to evaluate the comprehensive environmental quality status of the region,and the Seasonal Kendall test and Spearman correlation coefficient method were used to quantitatively analyze the change trend and correlation of water quality factors.The results showed that:1) From 2011 to 2018,the overall water quality of Xincun Port’s mariculture area was excellent,which could meet the functional requirements of the aquaculture area.2) The eutrophication status was good.Since 2011,it had been at an overall oligotrophic and mesotrophic level,and only became eutrophication in 2016.3) Water pollution was dominated by organic pollution,followed by phosphorus nutrient pollution,while heavy metal lead,arsenic pollution and pesticide residues in seafood also required attention.4) Pollution from ship navigation had increased and pollution from aquaculture had been reduced.By studying the changes of environmental quality in the sea area of Lingshui Xincun Port in the past eight years,we could provide scientific basis and intellectual support for the rational development,utilization and protection of Xincun Port.

    Studying on suitable light intensity,cultivation density and development degree for the gametophyte advance development of Saccharina japonica

    This study sought to investigate the optimum light intensity,cultivation density and the gametophyte advance development degree of Saccharina japonica,in order to provide technical support for gametophyte seedling.The suitable light intensity and cultivation density for the gametophyte advance development were determined by inspecting the female gametophyte development rate,ovulation rate and seminal vesicle formation rate.The suitable degree of gametophyte advance development was determined by inspecting the emergence rate and emergence time.It was found that the advance development speed of gametophyte under 37.5~62.5 μmol·m-2s-1 was significantly faster than that under 25.0 μmol·m-2s-1 in the condition of(13±1)℃ and 10.0 g·L-1.The advance development speed of gametophyte in the density of 5.0~10.0 g·L-1 was significantly faster than that in the density of 15.0~20.0 g·L-1 in the condition of(13±1)℃ and 37.5 μmol·m-2s-1,and only partial gametophyte developed,some gametophyte grew filament in the density of 15.0~20.0 g·L-1.The emergence test was carried out after the gametophyte was advance developed under the conditions of(13±1)℃,37.5 μmol·m-2s-1 and 10.0 g·L-1.The emergence rate were over 96.0% after 5 days of cultivation when the gametophyte was advance developed for 11~12 d,while the emergence rate was 90.7% after 9 days of cultivation when the gametophyte was without advance developed,which was 4 days later than the advance developed gametophyte.As a result,the suitable condition of light intensity and cultivation density were 37.5 μmol·m-2s-1 and 10.0 g·L-1 for gametophyte advance development of S.japonica,while the suitable advance development time was 11~12 d.

    Bangia fuscopurpurea is a kind of high-value economic seaweed rich in EPA and other fatty acids and it is a unique cultured algae in Fujian Province.In order to further promote the cultivation of B.fuscopurpurea and solve the industrial problems of high impurities and rotten food existing in traditional raft cultivation,we conducted the cultivation mode of suspension propagation in kelp phytotron glass room with temperature 9.0~22.0℃ during the period from December 9,2021 to February 10,2022.The results showed that the maximum average daily growth rate could reach 18.18%·d-1 and 21.39%·d-1 in the 50 L film bag air-lift photobioreactor and 1 000 L culture tank,respectively,which the yield was much higher than that in indoor cement pond culture and marine raft culture.Ash content and crude fat separately increased by 20.81% and 25.00%,protein content decreased by 18.15% compared with raft net curtain cultivation,at the same time,the quality of unsaturated fatty acids was also better than that the latter.Land-based suspension propagation aquaculture could effectively avoid the problem of impurity pollution in marine aquaculture which was of great value to improve the processing efficiency and quality of B.fuscopurpurea.In this paper,we reported the land-based aquaculture of B.fuscopurpurea for the first time,which will provide new models and technologies for the facility aquaculture of macroalgae.

    The feeding and the growth characteristic of larva and juvenile of Opsariichthys bidens were studied.The results showed that larva and juvenile of O.bidens were breeded under condition of water temperature 26~30℃.36.67% of the larva opened their mouth and started to feed after age of 3 days.The feeding rate of larva and juvenile was 100.00% after 5 days of age,the average rate of larva and juvenile in the full stomach was 99.20% after 6 days of age,and the relationship between feeding amount(y) and body weight(W) was calculated to be y= -0.000 4 W2+ 0.214 0W-0.200 8.The recommended dietary allowance of the larva and juvenile of O.bidens as 45%~50% of the body weight.The regression equation of total length(Lt) with day age(D) was expressed as Lt=-0.008 1D2+ 0.903 1D +4.159 5.The relationship between the body weight(Wt) and day age(D) was calculated to be W t= 0.075 0D2 +2.027 1D -10.689 0,and total length(Lt) and the body weight(Wt) could be expressed to be Wt= 0.001 3 L t 3.609 ? 8,b = 3.609 8,which showed that the growth of larva and juvenile of O.bidens belongs to the type of allometric.The results provided more references for the artificial breeding of O.bidens.

    Acute toxicity test of ammonia nitrogen to Litopenaeus annamei juveniles under low salinity and high alkalinity

    Under static water conditions with low salt and high alkalinity(salinity 5,total alkalinity 250 mg/L),the acute toxicity test was conducted to examine the effect of ammonia nitrogen on Litopenaeus annamei juveniles with body length of(1.12±0.45) cm.Results showed that median lethal concentrations(LC50) of ammonia nitrogen were 1 001.17,63.41,47.98,28.30 mg/L at the time point of 24,48,72,96 h respectively under low salt and high alkalinity conditions,so that its safe concentrations of total ammonia nitrogen and ionic ammonia nitrogen for Litopenaeus annamei juveniles were 2.83 mg/L and 0.22 mg/L,respectively.Ammonia nitrogen is highly toxic to Litopenaeus annamei,and measures to reduce the content of ammonia nitrogen have been proposed.

    Distoechodon compressus plays an important role in regulating and stabilizing the water ecosystem.In view of the decline of wild resources and its ecological water purification function,stock enhancement of Distoechodon compressus was carried out by Longyan City to restore its population in each main water area and achieve the expected effect.Based on the analysis of stock enhancement data from 2018 to 2021,the variety selection of stock enhancement and the artificial propagation of D.compressus were analyzed and discussed in this paper.Meanwhile,the key technologies,development and research direction of this field were also prospected,in order to provide reference for stock enhancement of D.compressus.

    Aquatic seed industry is the cornerstone of the development of aquaculture.Promoting the construction of commercial seed breeding system can solve the prominent problem of the separation between scientific research and market in the traditional breeding system.In order to further promote the development of aquatic commercial breeding,this paper analyzed the development status and shortcomings of China’s aquatic seed industry enterprises through literature query,data statistics and case comparison.It was considered that the development momentum of China’s aquatic seed industry enterprises was good,which was reflected in the improvement of enterprise breeding awareness,the effective application of seed conservation and control technology,and the diversity of commercial breeding forms.At the same time,China’s aquatic seed industry enterprises also had problems such as weak enterprise competitiveness,development potential and risk resistance,which were reflected in weak independent innovation ability,less investment in breeding R & D,small enterprise scale and so on.In this regard,we proposed to further enhance the competitiveness of China’s aquatic seed industry and promote the commercial development of aquatic breeding by formulating breeding innovation strategies from the two aspects of breeding objects and breeding process,extending vertically and broadening the seed industry chain horizontally.

    During the last decade of the 20th century,the artificial breeding of Larimichthys crocea accomplished its industrialization in China.Since then,the breeding scale and industrial division of the L.crocea industry have been continuously upgraded,forming a perfected industrial chain covering supplying the fishery necessities(fry,feeding stuff,facilities,etc.) and acquisition,trade,processing and export of the marketable fish.The supporting industries such as logistics,catering and tourism have also been gradually formed.In this report,industry experts and practitioners organized by the L.crocea branch of the China Fisheries Association conducted a comprehensive investigation of China’s L.crocea industry status in 2020,then summarized and analyzed it from ten aspects:seedling breeding,farm distribution and output,feeding stuff usage,major epidemic diseases,quality and safety,yields and market,Corona Virus Disease-19 impact,brand culture construction,supporting policies,financial and insurance services,and existing problems;gived countermeasures and suggestions on the industrial development for reference.The survey results showed that there were some problems in the development of L.crocea industry in China,such as low coverage rate of fine varieties,illegal drug use,low level of intensive and deep processing. Suggestions were put forward to promote the healthy and sustainable development of L.crocea industry in China by vigorously developing seed industry projects,standardizing industrial behaviors,increasing the proportion of medium-sized products,improving the processing rate,winning government support and creating industrial culture.

    The era of fishery big data has come.Some countries have started to carry out fishery big data research.Machine learning is an indispensable and effective means for big data research.Applying machine learning technology in fishery research is very necessary and important to make up for the deficiency of fishery big data analysis and application,and to develop the digital and intelligent fishery in China.The present situation and developing trend of applying machine learning to fishery research at home and abroad were studied by literature analysis.Based on the current research results of machine learning technology in fishery production,fishery classification,water quality detection,target recognition,fishery services and other fields,it was thought that machine learning had played an important role in fishery research.But machine learning was used for micro research more than macro research.And it was used for static research more than dynamic research.It was used even rarer to study interactively with other industries.At present,intelligent fishing products and some fishery observation cloud platforms had emerged.The technologies of Internet of Things also facilitated the widespread use of machine learning in fishery research.Strengthening the hardware infrastructure construction of Internet of Things,and strengthening the cooperation and integration with upstream data were the only approach for machine learning’s being widely applied in fishery research.Now,machine learning had a promising future in the research fields such as fishing activities’ supervision,fishery ecological simulation and producing a variety of daily necessities with fishery products.
