    Karyotype analysis of hybrid F1 groupers (Epinephelus akaara ♀ × E. moara ♂)

    In recent years, with the development of the grouper industry, hybrid breeding has played an important role in the development and cultivation of new species of grouper farming. The karyotype analysis of fish is of great significance in fish hybrid breeding, genetic relationship and genetic variation. Fisheries Research Institute of Fujian carried out research on the hybrid of Epinephelus akaara ♀ and E. moara ♂ in 2013, and successfully obtained the hybrid F1 (E. akaara ♀ × E. moara ♂), but no chromosome karyotype analysis of hybrid F1 has been reported. Thus, using E. akaara as female parent and E. moara as male parent, we obtained hybrid generation of normal development by artificial insemination technology, which called “Red cloud grouper”. In this study, the karyotype of F1 groupers (E. akaara ♀ × E. moara ♂) was studied. The juvenile fish were intraperitoneally injected with bovine serum albumin (BSA), followed by phytohemagglutinin (PHA) and colchicine solutions. The metaphase chromosome spreads were obtained from head kidney cells. The results of the study showed that the number of chromosomes of the hybrid F1 groupers (E. akaara ♀×E. moara ♂) was 48 (i.e. 2n=48), of which one pair was metacentric chromosomes (m), one pair was sub-metacentric chromosomes (sm), and two pairs were sub-telocentric chromosomes (st) and twenty pairs were telocentric chromosomes (t). Among these 48 chromosomes of the hybrid F1 groupers (E. akaara ♀×E. moara ♂), the largest relative length was (5.15±0.28), and the smallest relative length was (2.84±0.11). The number of chromosome arms (NF) of hybrid F1 groupers (E. akaara ♀×E. moara ♂) was 56. According to the analysis, karyotype formula of hybrid F1 groupers (E. akaara ♀×E. moara ♂) was 2n=48=2m+2sm+4st+ 40t, NF=56. So far, karyotypes of 29 species of groupers have been reported. The chromosome number of these groupers is 2n=48, and the chromosome number of the hybrid F1 groupers (E. akaara ♀×E. moara ♂) in this paper was consistent with this. It can be seen that 2n=48 may be the most basic karyotype feature of groupers. During the evolution of groupers, the number of chromosomes is highly conserved. However, the karyotypes of different species are significantly different. The same grouper species may also have different karyotypes obtained by different researchers due to different geographic regions, different chromosome preparation methods or operating errors. In this study, the number of chromosomes of the hybrid F1 of E. akaara ♀ and E. moara ♂ was the same as that of the two parents, but the chromosome karyotypes were quite different from those of the two parents. The chromosomes karyotypes could not all be found in the two parents. Maybe, a complex chromosomal mutation had occurred. This research may provide an important scientific basis for the genetic relationship analysis, genetic variation of the hybrid F1 groupers (E. akaara ♀ × E. moara ♂), as well as the germplasm identification, character selection and germplasm improvement of the hybrid offspring.
    The research on the influence of genetic diversity and growth of the Marsupenaeus japonicus progeny treated with 60Co-γ radiation

    The present study conducted four different radiation dose of 50 cGy, 100 cGy, 1 000 cGy and 1 500 cGy in order to research the new kind breeding of Marsupenaeus japonicus. In our study, the 60Co-γ radiation was conducted on the condition of 3.4 cGy/sec with M. japonicus which was about to spawn. We applied the AFLP molecular technology to analyze the genetic similarities, genetic distances and genetic diversity of the 60Co-γ-radiated progeny in each group. The weight data of M. japonicus progeny in each group grown in earth ponds over 90 days were also counted to supply the proof for the radiation breeding programme of M. japonicus. All those experimental data were conducted with single factor analysis of variance to test the difference. The experiment results indicated that the genetic similarities was from 0.873 7 to 0.894 7 and the genetic diversity of 60Co-γ-radiated progeny was not significant different from the control group in populations (P>0.05). But we found some obvious distinctions in the genome type of single individual in 60Co-γ-radiated groups compared to others which did not exist in control group. Meanwhile, the growth in earth ponds was significant different in each group. Our research provided fundamental data and science proof for the radiation breeding programme of M. japonicus.
    Isolation, identification and oxidation characteristics of marine sulfur-oxidizing strain Thiomicrorhabdus sp.B2-1

    In order to explore high-efficiency sulfur-oxidizing strains from mariculture environment, a marine sulfur-oxidizing bacterium, designated as strain B2-1, was isolated from the offshore sediment of culturing area of Meizhou Bay, Fujian Province. The morphological, physiological properties and 16S rRNA gene sequence, as well as the thiosulfate oxidizability under different culture conditions were analyzed. Strain B2-1was a Gram-negative, short rod bacteria and motile with a single polar flagellum.16S rRNA gene sequence analysis indicated that strain B2-1 was most closely related to the Thiomicrorhabdus indica 13-15AT (100% sequence similarity), thus designated as Thiomicrorhabdus sp. B2-1. The thiosulfate oxidation of strain B2-1 was observed at pH ranging from 6.0 to 9.0 (optimum, pH 8.0) and temperatures from 20 to 35°C (optimum, 30°C). The oxidation ability of strain B2-1 increased first and then inhibited with the increase of Mg2+concentration, with the optimum concentration of Mg2+ as 50 mmol/L. The addition of NH4+ could shorten the lag period and enhanced the oxidation ability. The oxidation rate could raise from 47.4% to 95.5% with the addition of 187 mmol/L NH4+(P<0.05). Thiomicrorhabdus sp. B2-1 has great application potential for sulfur oxidation in mariculture environment control.
    Design and preliminary experiment of floating auto-feeding system for net cage culture in the bottom waters outside the bay

    Abstract: In recent years, with the construction of various large-scale aquaculture facilities at home and abroad, it has opened the prelude for the aquaculture industry to enter the deep sea and expand the aquaculture space outside the bay. In response to trends to promote the transformation and upgrading of the aquaculture industry, the author’s research team is developing large-scale lifting rigid cages that are suitable for the bottom waters outside the bay. In order to solve the problems of feeding management in the net cages culture, we have developed a floating auto-feeding system. The system uses built-in batteries to provide the energy, and offshore automatically realizes a series of precise actions such as timing, fixed-point, and quantitative feeding of bait with the help of a built-in PLC control program. At present, the miniaturized prototype of the system has been developed and preliminary experiment has been carried out. The total volume of the feed bin is 0.23 m3, and the total volume of feed storage is 164 kg. The four independently controlled feed bins can realize to feed at different intervals. The average feeding ability is about 66.09 kg?min-1 (size of bin door opening: 90 mm×100 mm). The preliminary experiment shows that the floating auto-feeding system basically meets the design requirements. It has the advantages of simple structure, strong performance in adapting to harsh sea conditions, convenient operation, accurate feeding action, low failure rate, and stable operation, etc.
    Optimization of the recovery of protein from washing solution of the Fugu surimi by FeCl3 flocculation

    Rinse is one of the important steps of minced fish processing. Lots of soluble protein would be lost in the process of rinsing. So, recovering protein from the minced fish rinse solution can improve the utilization rate of the surimi, reduce the cost of sewage treatment steps, and decrease the pollution of environment. In this paper, FeCl3 as a flocculant was used to retrieve the water soluble protein from the surimi rinse water. According to different single factor variables, the optimal conditions of the recovery of protein from the surimi rinse water by FeCl3 flocculation method were researched and optimized by response surface analysis. Results showed that the optimal conditions of FeCl3 recovery proteins were: pH 6, temperature of 65℃, and an addition amount of 1% FeCl3 solution of 1.4 mL in 10 mL minced fish rinse solution.
    Optimization of extraction conditions and investigation of the antioxidant activity of polysaccharides from Bangia fusco-purpurea

    In the present study, the extraction process of polysaccharide from Bangia fusco-purpurea was optimized and the antioxidant activity of polysaccharide was studied in vitro. The effects of the extraction time, extraction temperature and ratio of material to liquid on the extraction amount of polysaccharide from B. fusco-purpurea were investigated, respectively, and the optimum preparation conditions were selected with orthogonal design combined with multi-index test. Meanwhile, the antioxidant activity of polysaccharide from B. fusco-purpurea was evaluated by DPPH free radical and hydroxyl radical scavenging method. The results showed that the optimum extraction conditions were the ratio of solid to liquid was 1∶40 (m/v) at 90℃ for 2.0 h. Under the optimum extraction conditions, the maximum extraction yield of polysaccharide reached (77.57±2.17) mg/g. Additionally, the result of antioxidant activity in vitro showed that the polysaccharide form B. fusco-purpurea presented significant scavenging effects on DPPH free radical and hydroxyl free radical. The IC50 value of DPPH free radicals scavenging was 0.36 mg/mL, and the IC50 value of hydroxyl free radicals scavenging was 0.65 mg/mL. This study can be used for the further development and application of B. fusco-purpurea polysaccharide.
    Establishment and implementation of HACCP system in canned oil smoked scallop enterprises

    In order to ensure the quality and safety of canned oil smoked scallops, HACCP principle was applied to analyze the hazards in the whole process from raw materials to consumption. Five key control points including raw material acceptance, can filling, can sealing, sterilization and cooling were determined. The critical limits were set at the critical control points to monitor, record and correct errors. And then, the GMP and SSOP were systematically analyzed as the basic systems to establish the HACCP system, and the objects to verify whether the HACCP system operates effectively in the implementation process were discussed from two aspects of note audit and on-site audit, so as to ensure the preciseness and effectiveness of HACCP system.

    From August to September 2018, 17 oyster samples were selected from 17 major shellfish cultivation areas in 6 coastal cities of Fujian Province. The investigation found that the concentration of Cu and PHCs in oyster samples from some cultured sea areas exceeded the second class marine biological quality standard. The correlation analysis showed that there was a strong positive correlation between Cu and Cd, Pb and PHCs, BHC and PCBs, as well as Cu and PCBs. Through the principal component analysis of the test parameters, the variance contribution rates of the three principal component factors were 29.436%, 24.938% and 15.838% respectively, and the cumulative contribution rate reached 70.211%. Cu, Hg and PCBs were the main driving factors of primary pollutants in oysters from Fujian coast.

    Under the condition of continuous administration, the present study investigated the metabolic residue of enrofloxacin and its metabolite ciprofloxacin in (Epinephelus fuscoguttatus ♀ × Epinephelus lanceolatus ♂), in order to provide theoretical basis for the scientific and reasonable use of enrofloxacin to prevent and treat the bacterial diseases of (E. fuscoguttatus ♀ × E. lanceolatus ♂), and to provide reference for the guidance of enrofloxacin usage in aquaculture. (E. fuscoguttatus ♀ × E. lanceolatus ♂) weighted (300±28) g were fed with 30 mg/(kg?bw) enrofloxacin by oral administration under the water temperature of 28℃, then the concentration of enrofloxacin and its metabolites ciprofloxacin in plasma, muscle and liver were determined by HPLC-MS. The results showed that enrofloxacin quickly reached the maximum in plasma, liver and muscle, while the peak time was 3 hours at the end of the fifth day, 3 hours at the end of the fifth day and 3 hours at the end of the fourth day, respectively. The concentrations were 24.5 mg/L, 13.0 mg/kg and 8.20 mg/kg, respectively. The peak time of ciprofloxacin in plasma, liver and muscle was 3 hours after the end of the fifth day, 3 hours after the end of the fifth day and 3 hours after the end of the fourth day, and the concentrations were 0.276 mg/L, 0.738 mg/kg and 0.110 mg/kg, respectively. Our results indicated that only a small amount of enrofloxacin was metabolized to ciprofloxacin, and it is suggested that the rest period should not be less than 980 degree-days.
    Current situation and suggestions on the exploitation of Jack mackerel resources in the world

    Jack mackerel is one of the important pelagic fishes and has become an important part of the world marine fishing industry. Based on the production statistics of FAO from 1970 to 2018 and relevant literatures, the present situation of the exploitation of jack mackerel resources in the world was analyzed and some suggestions were put forward. The study found that between 1970 and 1995, the world production of jack mackerel showed a stable growth trend, reaching an all-time high of 7.91 million tons in 1995, followed by a significant decline. Jack mackerel production during 2010—2018 was at a low level, between 2.97 and 3.33 million tons, accounting for 3.64%~4.12% of the total catch. The highest annual yield of more than 10×104 tonnes was recorded for Trachurus murphyi, Trachurus trachurus, Trachurus capensi, Trachurus japonicas, Trachurus trecae, Trachurus declivis, Trachurus mediterraneus, and Trachurus indicus,et al. In order to promote the sustainable development of the world's fishing industry of jack mackerel, the study made the following recommendations: Development of new fishing grounds and resources for jack mackerel; International cooperation, including through international fisheries organizations, on climate change and resource volatility, as well as resource assessment and management strategies for jack mackerel, to improve resource forecasting capabilities.
    Thoughts on informatization construction of coastal fishing ports in China

    The fishing ports have played an important role in the economic and social development of fisheries and the adjustment of the industrial structure, where information technologies have provided new ideas for fishing port supervision, fishing vessel management, marine catches tracing, and environmental governance. In the present study, the research on the status of fishing port informatization included two aspects. One aspect was the current situation of fishing port information systems, which were classified into five areas, namely, video application, fishing vessel monitoring application, information management application, decision support application and government service application. The other aspect was the distribution of data resources and the problem analysis which summarized as insufficient data analysis, difficult in data collection, data inconsistent and uneven data distribution. A framework for fishing port information system, which including the bottom-up structure of information infrastructure, data management, technology stack and fishing port business, and information standard and safety as guaranteed, was established with the overall objective of sustainable development of the fishing port economy. Based on current fishing port information infrastructure, three stages of key technologies of fishing port informatization were proposed. First, as the next research direction of fishing port video monitoring system, the fishing port ultra-large-scale vision system brought challenges in data acquisition, processing, transmission, analysis and hardware stability due to the large number of fishing ports, huge amounts of data, rapid data changes, and the complex ecological environment of fishing ports. Second, the Fishing port cloud-end collaborative integration technology would build the intelligent perception of fishing port terminals and the rich computing resources in fishing port cloud. Third, the Single-multibody-ecological fishing port intelligent networking technology would provide applications such as overall perception of fishing port, interactive intelligence in fishing ports groups, intelligent scenes of fishing ports, collaborative intelligence between fishing ports and fishing vessel and ecological intelligence of fishing port groups. The research in this paper provided a reference basis for the planning and construction and management decision of the fishing port informatization in the next step. The future of fishing ports was not only the investment and construction of information hardware facilities, but also innovations in information applications. By constructing a fishing port cloud-end architecture and using data analysis as the core brain, it would create fishing port production management, fishing port economic zone construction, fishing port global planning, and more applications in multiple fishing port business areas.
    A general situation of fishery resources enhancement in Haiyang City of Shandong Province

    Based on the data of fishery resources enhancement in Haiyang City of Shandong Province, this paper had a general situation for the fishery resources enhancement work over the recent twenty years. After years of development of the fishery resources enhancement, the species of releasing from one to eight and the number of releasing from 30 million units to 725 million units. The species included Penaeus chinensis, Penaeus japonicus, Portunus trituberculatus, Rhopilema esculentum, Acanthopagrus schlegelii, Sebastes schlegelii, Paralichthys olivaceus, Sepia esculenta, Sepiella maindroni. The population resources had got the recovering of certain level after enhancement. The average annual output value had reached 39.96 million yuan, 20.28 million yuan, 41.74 million yuan of Rhopilema esculentum, Penaeus chinensis, Portunus trituberculatus. The fishery resources enhancement also stimulated the development of five nursery enterprises and sixty Fishery Enterprises. These companies had created lots of jobs that stimulated more than 40 000 people to work. The fishery resources enhancement had a good results and produce significant the ecological, economic and social benefits. In the end, the paper put forword some suggestions to improve the fishery resources enhancement which to provide references and bases for the effective implementation and scientific management in the future.
    Research progress of oyster polyploid

    Oyster tastes delicious and is rich in nutrients such as zinc, taurine and glycogen. With rapid growth, high yield, short food chain and good economic benefits, oyster has become an important shellfish for aquaculture in China and even the world.The main cultivated oyster varieties in China are Crassostrea gigas (also known as Pacific oyster), Fujian oyster (also known as Portuguese oyster), C. ngulata, C. hongkongensis, C. riakensis, C. sikamea, etc.At present, the phenomenon of smaller individuals, slow growth rate, longer growth cycle and decreased quality in the cultivation practice is in urgent need of strengthening the selection and breeding of oyster varieties, and the development of polyploid breeding technology can effectively promote the development of oyster industry to the direction of high quality and high quantity.The main methods of oyster triploid induction include biological hybridization between diploid and tetraploid, physical induction methods such as temperature shock, hydrostatic pressure, electroshock and osmotic pressure, and chemical induction methods such as CB, 6-DMAP and caffeine.This review is expected to provide reference for the study of polyploidy in oysters and other shellfish.
