Under different hatchery conditions, effects of diet and environment on growth and survival of the sea cucumber Apostichopus japonicus larvae were investigated for 7 days by feeding oceanic red yeast or shading net covering. The results showed that the survival rates of the cage hatchery group were over 52.4%~57.9%, which is lower than the indoor hatchery group. When feeding with oceanic red yeast, the larvae growth rate was 86.4 μm/d, the survival rate was 55.1%±8.0%. When feeding with no diets, the growth rate was 85.1 μm/d, the survival rate was 53.0%±7.5%. No significant difference was found between fed and unfed group. The larvae survival rate in net covering group was 52.8%±7.4%, which was significant higher than the survival rate 46.9%±7.0% in uncovering group. These results should be valuable to direct the technique optimization of artificial breeding for the hatchery production of A. japonicus larvae.

When enrofloxacin residue is close to the maximum residue limit in aquatic products, it is very important to carry out evaluation of measurement uncertainty to accurately determine whether the products meet the quality standards. So a method of uncertainty evaluation for the determination of enrofloxacin residue in aquatic products by liquid chromatography was established. According to Evaluation and Expression of Uncertainty in Measurement (JJF 1059.1— 2012) and Guidance on Evaluating the Uncertainty in Chemical Analysis (CNAS—GL06: 2006), a mathematical model was constructed and the uncertainty sources were analyzed, and then the main components of uncertainty were quantified and combined. Finally the expanded uncertainty was achieved. When the measured value of enrofloxacin residue in aquatic products was 97.1 μg/kg, its expanded uncertainty was 5.6 μg/kg. The results showed that the major factors of uncertainty were sample pre-treatment and standard solution. The uncertainties of sample pretreatment and standard solution accounted for more than 50% and 22% of the total uncertainties, respectively.

Seven strains of bacteria were isolated from the diseased eel, and which were identified as Vibrio vulnificus through 21 kinds of biochemical indexes analysis. The specific primers amplifying hemolysin gene (vlly) of Vibrio vulnificus were designed to detect the isolated bacteria genome DNA, the PCR results indicated that the specific fragments of 352 bp DNA could be amplified from the isolated Vibrio vulnificus, but could not amplify any positive band from non-Vibrio vulnificus bacteria. The mouse serum anti the O antigen of Vibrio vulnificus were prepared, and the assay of agglutination reaction showed that the serum anti- O antigen of Vibrio vulnificus from eel could not agglutinate to the strain 1.1758 of Vibrio vulnificus isolated from human. However, the serum anti O antigen of FJ03-X2 could agglutinate to the most strains of Vibrio vulnificus from Anguilla anguilla. These results suggested that the PCR primers based on hemolysin gene could specifically detect the strains of Vibrio vulnificus, the serovar of Vibrio vulnificus from eel was different from other sources, and the strain of FJ03-X2 had good immunogenicity O antigen and could be as candidate strain for vaccine development of Vibrio vulnificus.
