    Analysis of Endosulfan and Its Metabolite Residues in Sediment From Fisheries Environment by Gas Chromatography

    A method was developed for the determining of endosulfan and its metabolite in sediment from fisheries environment by gas chromatography. The samples were extracted with n-hexane/acetone(V/V, 1:1), sulfide were removed by copper powder, and purified on a carbon-florisil SPE column eluted with dichloromethane/n-hexane/acetone(V/V, 3:7), then analyzed by gas chromatography with electron capture detector(GC-ECD) using the external standard. The linear range of each endosulfan derivative were 0.001~0.200μg/mL (r≥0.9996). When spiked with 1μg/kg, 10μg/kg, 20μg/kg , the average recoveries ranged from 76.3% to 107% and their relative standard deviations(RSD)were between 2.31% and 11.8%. The limits of detection (LOD) of α-endosulfan, β-endosulfan and endosulfan sulfate were 0.03, 0.04, 0.05μg/kg(dry weight) respectively, while the limit of quantization (LOQ) was 1μg/kg (dry weight) for all of them. This method can be applied in the detection of endosulfan contamination in fisheries environment.
    Identification and Antibiotic sensitivity of Aeromonas hydrophila Isolated from Skin ulcer of Artificial breeding Anguillaanguilla

    One Aeromonas hydrophila strain was isolated from the sick artificial breeding Anguilla anguilla from FuJian ZhaoAn. The isolated could induce symptom of natural disease in Anguilla anguilla, have β-hemolytic, could grow in the salt content at 4%, gram's staining negative, short rods, athletically inclined. Biological characterization showed that the isolated was similar to A.hydrophila isolated in other aquatic anima1. Analysis of 16S rRNA gene showed that the isolate shared more than 99.8% identity with A.Hydrophila. Based on morphological, biochemical and phylogenetic analysis, the isolated was identified as A.Hydrophila. The antibiotic sensitivity test showed that the isolated bacteria was hypersensitive to Cefotaxime Sodium, Florfenicol, Compound Sulfamethoxazole, Ofloxacin, Cefepime and Cefamandole.
    Present situation Investigation of Zooplankton near Yueqing Bay

    The Zooplankton species composition,quantity distribution,diversity,community structure and relationship between abundance and environmental factors in Yueqing Bay based on the survey in May and September. The results showed that there were 13 groups 47 species of Zooplankton in May and 16 groups 70 species in September. The Zooplankton in the Survey area were mainly composed of coastal low saline species, and there were a few of brackish water species. The main species of May were Calanus sinicu、leurobrachia globosa、Sagitta bedoti Beraneck, and the main species of September were Centropages dorsispinatus、Sagitta nagae、Calanopia thompsoni、Euphausiacea larvae and Macruran Larvae. The abundances of zooplankton in May were 91ind /m3, and 115ind /m3 in September. The qualities of of zooplankton in May were 0.209 g/m3,and 0.222 g/m3 in September. The Margalef richness index、 Pielou evenness index、Shannon-Weiner diversity index and Simpson dominance index in September was more than in May. Through the study of the Zooplankton species composition、quantity distribution、diversity、community structure and relationship between abundance and environmental factors in Yueqing Bay is to provide evidence for ecological monitoring, evaluation, and provide scientific evidence for the development of the sustainable utilization of marine resources to the sea.

    Abstract: This paper analysed and discussed the process of a Prorocentrum donghaiense bloom, which occured in Changle Sea Zone of Fujian Province during May 2 and May 10, 2010, based on continuous monitoring of it. This red tide organism could form red tide under weather condition of intense illumination; this organism mainly utilizes nitrate as its nitrogen nutrient after the formation , and it also has remarkable ability to withstand the stress of low phosphorus in the environment. In addition, the principal components analysis of environmental factors in Changle sea area during the monitoring process showed that limiting factors of this red tide include NO3-N, NO2-N, water temperature, salinity, et al.

    Abstract: A red tide in Tong’an bay, Xiamen Seas during July 26 to August 7 2011 was reported in this paper, and the in situ hydrological, meteorological, chemical and biological factors were analyzed. It was indicated that this red tide with the area of 105 km2 was a hybrid triphase red tide in which the red tide species include Skeletonema costatum, Akashiwo sanguinea, Chaetoceros sp.. The major causes were eutrophication, continuous high air temperature and the water body stability of neap tide, and the air pressure due to “Luotan” typhoon and sharp decline of salinity due to rainfall also contributed to this red tide

    In order to understand the Fuqing Bay surface sediments, heavy metals pollution status and its potential ecological hazards, as well as heavy metals and organic matter content in the correlation, from June to August in 2009 we investigated Fuqing Bay surface sediments, heavy metals and organic matter content, and used single factor index method, and Hakanson potential ecological risk index to evaluate it comprehensively. The results show that, Fuqing Bay surface sediments of Cu, Pb, ,Cd, Hg, As and organic matters, the average content are 17.4 × 10-6,31.7 × 10-6,0.058 × 10-6,0.079 × 10-6,8.90 × 10 -6 and 1.31%, which are consistent with a type of marine sediment quality standards that means it has good quality. Apositive correlation of heavy metals, especially a positive correlation with organic matter is obvious. Heavymetal single factor index in decreasing order is: Pb (1.27) > As (0.59)> Cu (0.58)> Hg (0.40)> Cd (0.12), heavy metal pollution index was 2.96 for the low-pollution level; individual potential ecological risk of heavy metals parameters were Hg (15.8)> Pb (6.35)> As (5.94)> Cd (3.48)> Cu (2.91), the potential ecological risk index (RI) was 34.5. which indicated that Fuqing Bay existed a low potential ecological risk.

    Abstract: In June 22nd 2004, our institution was entrusted by the local fishery management station to investigate the large area death accident of four major Chinese carps in the fishery farm at Gutian Reservoir in Fujian, China. We collected water and fish samples from 5 stations in accident area. By comparing the water and fish organs samples from the control site, we basically excluded the pollution causes such as acid-base chemicals, hypoxia, heavy metals, fecal (not ammonium poisoning), fluoride, volatile phenol and water quality (elevated sulphide and bacterial). Our investigation result shows that the accident was caused by the pollution of one organic chloride pesticide: endosulfan, and it is a intentional poisoning action. As environmental pollution indicating organisms, plankton and microorganisms were proved to be essential for such investigation. Water quality was indicated by the change of species structure, abundant and morphology of phytoplankton and zooplankton. We also learned that it was helpful for our investigation that the fish farm owner could reserve the dead sample (freezed) and report to the local fishery management department in time. The fishery administrative departments and agencies in the fishery after receiving the report shall timely notify the fishery environment monitoring and other relevant qualification technical units in accident sample identification, in order to ensure the effective handling of fishery pollution accidents, help farmers to retrieve the economic loss, preserve the fishery ecology environment.

    Abstract: The total DNA yield in the cell nucleus vary with different species, the species DNA ploidy can be determined according to the analysis of the DNA content of the cell by Flow Cytometry. In the present study, the DNA ploidy determination for hybrid abalone(♀Haliotis discus hannai ×♂Haliotis discus discus)was studied, cells suspension was provided from small tentacles of hybrid abalone, then conducted staining by Destination Access Point Identifier(DAPI), finally, DNA ploidy determination was finished by PA-Ⅱploidy analyzer. The results showed that the optimal time for oscillation was 1.5~2.0 minutes, the optimal staining time is 4.0~5.0 minutes.

    Wild Erythroculter ilishaeformis fry in Minjiang river basin were taken as experimental material , which fed on artificial formula feed after feed domestication. After three years of net cage culture in reservoirs, the wild fish completely adapted to the living environment of net cage; the mean weight reaching 1.2kg, parent Erythroculter ilishaeformis with sexual maturity and reproductive capacity were attained. In 2005 and 2006 the selected parent Erythroculter ilishaeformis cultured by ourself were used for conducting experiments of artificial breeding in June after two months of intensive cultivation. The mixture of LRH-A2 and HCG was used as artificial injection oxytocin; by natural spawning, fertilization, hatching, and fry rearing, a total of 1602000 Erythroculter ilishaeformis fry were attained.
    The Exploration of Controllable Ecological Aquiculture Technology in Breeding Pesudosciaena crocea Seedlings Inland

    Pseudosciaena crocea seedlings were cultured inland with controllable ecological aquiculture technology for 166 days.The results show that, the average weight of Pseudosciaena crocea raised from 0.1 to 40g,while the average lengh raised from 1.5cm to 16.5cm,and the food coefficients reached 0.97 while the survival rate was 44.1%. zero-water-exchange and zero-drug-use culture was achieved in the process of the trial.

    With increasing serious situation of food supervision recently, the detection technology of qualitative safety of domestic aquatic products has improved significantly, which requires more labor work and higher level of laboratory management. The traditional laboratory faces difficulty in efficiency, logistical management, Statistical analysis and tracking sample status. Laboratory Information Management Systems(LIMS)makes the whole laboratory environment as a management object, it uses computer network technology to improve the comprehensive laboratory management, which could improve laboratory efficiency, work level and data quality, and reduce the cost. At present, the system on aquatic product quality inspection industry is still at the initial stage. This article uses the aquatic product quality inspection laboratory to analyze the design idea and system architecture of LIMS, and discuss how to apply LIMS to laboratory analysis and management, and it also discusses difficulties of LIMS during the construction and in the process of research and experience.
