    Status and suggestions of mud crab aquaculture in Fujian Province

    The yearly output of the mud crab in Fujian Province ranks second in China recently. Mud crab aquaculture has developed into one of the key fisheries in Fujian Province. The main objective of this article is to present the current developing status of mud crab aquaculture in Fujian Province, including growout, larval rearing, disease control, genetic breeding and so on. Furthermore, the main problems affecting crab fisheries are analyzed and the concrete suggestions are proposed.
    Genetic Structure and Population Differentiation of Sipunculus nudus in China based on sequence analyses of mitochondrial COI gene

    Genetic structure and population differentiation in wild populations of Sipunculus nudus were researched in this paper. 91samples collected from Changle、Gulei、Jinjiang in Fujian province and 15 from Xiamen、Beihai、Sanya were studied based on sequence analyses of the mitochondrial cytochrome oxidase I(mtDNA COI) gene segment. 193 nucleotide polymorphisms and 52 haplotypes were found in total 106 samples. Among three Fujian geographical groups, gene diversities (π) ranged from 0.00155 to 0.01325, and the nucleotide diversities (h) from 0.626 to 0.972, showed that Sipunculus nudus has high genetic diversity. AMOVA analysis disclosed that variation between populations accounting for 98.03% of total variation, revealing that variations occurred between populations were the main source of the total variation. Neighbor-joining tree based on mtDNA COI sequence showed A、B、C and D four different branches with high support value. The four distinct clades among south China coastal waters revealed underestimate cryptic diversity within Sipunculus nudus. Minimum spanning network from all haplotypes reveals two distinct branches as Fujian and the northern Gulf in groups A, which may be approached to subspecies level upon further research. This study may be useful for the resources protection, artificial seed stocking and genetic breeding of Sipunculus nudus.
    Change of biochemical component content of tissue and organ during the development of spermary of Marsupenaeus japonic

    In this study, enzymatic, field sampling and other methods were used to study pancreatic morphology and several biochemical component content changes in spermary and liver of Marsupenaeus japonicus during the spermary development. It intended to illustrate in the natural state, the growth and decline and transfer condition of nutrition substances in the tissues and organs of Marsupenaeus japonica broodstock in reproductive process The results showed that in the natural state of the Taiwan Strait waters, Marsupenaeus japonicus, testis index from the phase I 0.010 ± 0.001~IV period of 0.017 ± 0.002 was gradually increased (P<0.05); liver and pancreas fell from Phase I of 0.021 ± 0.002~to stage IV 0.025 ± 0.003, while also showed increasing trend, but little change (P>0.05). Glucose, cholesterol and triglyceride levels (mg/dl) changed as follows: In the testis, is 14.85 ± 5.40 ~ 4.75 ± 3.17,7.69 ± 2.47~16.46 ± 6.79 and 15.85 ± 4.56~36.78 ± 8.12; in the liver pancreas, is 10.95 ± 2.60~22.24 ± 5.76,27.12 ± 9.11~202.62 ± 77.26 and 42.87 ± 13.39 ~ 116.99 ± 24.57; in the hemolymph, is 6.38 ± 1.44~11.25 ± 3.08,53.77 ± 32.10~155.32 ± 62.55 and 13.67 ± 7.06 ~79.56 ± 38.19. This paper also showed that during the development of Marsupenaeus japonicus spermary, how gonad、pancreas and liver changeed in morphological area. The situations of rise and fall with the transfer in liver, pancreas, blood, lymph, testis glucose, cholesterol and triglyceride were also discussed in detail.

    Abstract:In the winter of 2011 , the sea cage cultured sea cucumber suffered from serious disease in many sea area of Ningde, Fujian Province, accompanied by the phenomenon of death. The symptom displayed as anorexia, viscera ejection, general atrophy, and skin ulceration, the disease was tentatively termed as “skin ulcerative syndrome”. The bacterium CS1 and CS2 were isolated from the focal area of the infected sea cucumber, and they were dominant amongst the isolates. Artificial infection test proved that they were the causative pathogen associated with the disease. In addition, a lot of Boveria sp. was found from the respiratory tree of sick sea cucumber. The bacterium CS1 and CS2 were identified as Vibrio splendidus and Pseudoalteromonas, respectively, by the means of morphological , biochemical tests , and 16S rDNA sequence analysis. This paper revealed for the fisrst time that the causative pathogen leading to the“skin ulcerative syndrome”of the sea cucumber in the area, which will be helpful in the disease control and health management during sea cucumber cultivation.

    Response surface analysis was applied to optimize microcapsule technical of Squid liver oil. On the basis of single factor experiment, proportion of wall material, concentration of emulsion and temperature of into wind were chosen as response factors based on microcapsule embedding rate to establish regression model. The results show that model can prediction microcapsule effect of squid liver oil , and the optimum condition was obtained as follows: proportion of wall material (HI-CAP100∶cyclodextrins) of 92:23,concentration of emulsion of 21%,and temperature of into wind of 171℃, microcapsule embedding rate of 94.09% under this condition.
    Effects of transglutaminase on the quality of surimi products

    In this paper, the transglutaminase (TGase)was used to improve the quality of surimi products. The effects of TGase on the elasticity and cohesiveness of surimi products were tested at different TGase addition levels, reaction temperatures and time. Through the L9(34) orthogonal test,the experimental results showed that the quality of the surimi products was improved to a certain degree. The optimal parameters namely 0.10% TGase addition, reation at 35℃ for 1.5h were set.

    Abstract: Response surface analysis was applied to optimize the decalcification technique of Tilapia mossambica fish scale. Hydrochloric acid concentration, substrate concentration, reaction temperature and reaction time were chosen as the impact factors based on decalcification rate of fish scale. The appropriate levels and factors were selected by previous single factor experiments. According to the Box-Behnken central composite design, in accordance with the dynamic equation model, and significance of all factors and interactions among them were analyzed. The optimum condition of decalcification technique of Tilapia mossambica fish scale was obtained as follows: hydrochloric acid concentration of 2.9%, substrate concentration of 6.3%, reaction time of 1.9 h, and the decalcification rate of fish scale could reach 99.55% under this condition.

    Abstract: According to the shrimp survey data of four seasonal months in the northeast Fujian outer-sea (26o00′~28o00′N,120o00′~125o30′E) in May,August,November of 2008 and February of 2009,this paper studied and analyzed the quantitative distribution and biology of Trachypenaeus curvirostris. The results showed that the quantitative distribution was obvious different in the seasons and the waters, August were more and the ratio of its catch in total crabs catch was 60.2%, the average density of its resource was 614.5g/h and 144.7ind/h, May were less. The area of high biomass density occurs in the waters at depth of with 60m, the ratio of its catch in total crabs catch was 65.8%, and the average density of its resource was 1119.2g/h and 214.7ind/h; The body length varied between 47 and 120 mm, the dominant were between 60 to 85mm occupying 68.6%; The female was obvious larger than the male with the sex ratio 1:0.42, the average body weight was 6.4g and 3.2g respectively;The mature degree of the female was dominated by Ⅱ and Ⅲ stage,accounting for 44.7% and 43.8%;The feeding intensity was middling; The average resources capacity was 684t.

    Abstract: A growth experiment was conducted to evaluate the effects of various size range of marine sand on the growth and surviva1 rate of Sipunculus nudus L. in intermediate culture stage in 2010 and 2011.The results showed that the larva of Sipunculus nudus L. have a growth advantage and a higher surviva1 rate cultured in marine sand of 250um~500um than those which cultured in marine sand of 63um~250um.The results indicated that the size of marine sand wase important to improve Sipunculus nudus L’S intermediate culture.

    In this experiment, the seaweed juice processing method and the optimal formula of of seaweed juice Noodle was studied. The results showed that the best craft conditions were 2.5% of the soaking liquid concentration, the temperature at 80℃ and the time was 30min,we can make the flavourless seaweed juice.Wheat flours, flavourless seaweed juice , NaCl and Na2CO3 were mixed in the ratio of 100:80:3.7:0.2, We can made the better seaweed juice Noodle with Seafood flavor.

    Abstract:Effect of different feed on activities of digestive enzymes in Epinephelus coioides were examined in this articel.1500 Epinephelus coioides were randomly divided into three groups, each group of two replicates, using three different feed fed 50d, five Epinephelus coioides from each parallel were randomly selected to detect digestive enzyme activity. The results showed that different feed including the type and feed ingredients had significant effect on the digestive enzymes of Epinephelus coioides. In this experiment, feed No.2 had the highest amylase, feed No.3 had the highest overall material, fat digest mainly in the intestine, three lipase activities in the intestine was not significant.

    A growth experiment was conducted to determine the optimum feeding rate and the effects of different feeding rate on the growth performance of juvenil Takifugu obscurus Abe were studied. Four daily feeding rates were designed at 3%, 4%, 5% and 6% of the total body weight. Under the water temperature of 23℃, feed the fish that the average weight was 3.45-3.56g.After 30d growth experiment,the results showed that the relative weight gain, specific growth rate ,food conversion efficiency and survival rate were significantly (p<0.05) affected by feeding rate. the optimum feeding rate of juvenile Takifugu obscurus Abe should be 4%, on the feeding rate the relative weight gain was 116.04±0.04%,food conversion efficiency is 83.70±0.02%,specific growth rate is 111.49±0.03%/d,survival rat is 84.44±0.02 %.
