    In the present study, four sets of five specific primers were designed based on the conservative sequence of WSSV genome using PrimerExplorer V4. Five specific primers including one set of inner primers FIP and BIP, one set of outer primers F3 and B3, and one loop primer LF or LB. A visual WSSV-LAMP was developed in which calcein was applied to be detection indicator. Its detection limit for WSSV was determined by detecting the ten-fold diluted recombinant plasmid of pMD18-WSSV. The reaction temperature and time of WSSV-LAMP were at 63 ℃ for 60 min, respectively. Detection limit of the LAMP method for the WSSV DNA in which WSSV-2, WSSV-3 and WSSV-4 were determined to be 2.25 ~ 0.225 copies/μL, 2.25 ~ 0.225 copies/μL and 225 ~ 22.5 copies/μL, respectively. The LAMP assay could be finished within an hour, requiring only a regular laboratory water bath or heat block for reaction and the result could easily be detected using visual observation. Therefore, it is very suitable for detection in the fields that would provide a rapid detection approach for early WSSV infection in aquaculture.

    Nervous necrosis virus (NNV) can cause massive mortality in larval and juvenile stages of orange-spotted grouper (Epinephelus coioides). Due to the lack of viral genomes, virus-like particles (VLPs) are considered as one of the safest and most effective vaccine to prevent viral diseases. In this study, the expressions of three immune-related genes (IFN, TNF-α and IRF3) were analyzed in 9 tissues (heart, liver, spleen, kidney, intestine, sputum, blood and eye) of healthy groupers. The gene expressions in main immune tissues (liver, spleen and kidney) were also detected after vaccination by intramuscular injection. Results showed that mRNA transcripts of IFN, TNF-α and IRF3 were observed in nine tissues of the grouper, and the highest expression values of IFN, TNF-α and IRF3 were detected in brain, gill and blood, respectively. The vaccine alone was not able to enhance the expression of genes, including IFN, TNF-α and IRF3 in liver as well as IFN in spleen and kidney. But the expressions of these genes were up-regulated by injecting vaccine with CpG oligodeoxynucleotide (ODN) motifs. Vaccine immunity alone could up-regulate the expressions of IFN, TNF-α and IRF3 in other tissues, but their highest expression values were smaller than that of the corresponding adjuvant group, indicating that the efficacy of vaccine could be further enhanced by CpG ODN after vaccination.

    Isolation, Identification and virulence-related gene analysis of a pathogenic Vibrio alginolyticus from large yellow croaker

    Vibriosis is the most prevalent and the most harmful marine disease of marine aquaculture animals. large yellow croaker (Pseudosciaena crocea) , the The largest aquaculture fish is the main infection of Vibrio. Body surface ulcers, hemorrhage, and silk rot are the main symptoms of vibriosis. In this study, a dominant strain was isolated from the diseased large yellow croaker of the Ningde Jiaocheng Dawan fish raft of China, which had surface and fin ulcer, hemorrhage and gill defect. In order to identify the strain and evaluate its pathogenicity, the culture was purified and the physiological and biochemical characteristics of the strain were analyzed. The 16SrDNA of the strain was identified, the pathogenicity of the strain to the large yellow croaker was determined, and the virulence factor of the strain was also tested. The culture results showed that the strain could grow on TCBS medium, and the colony color was yellow. The physiological and biochemical identification results indicated that the strain was Gram-negative Brevibacterium, which was halophilic, and the physiological and biochemical indexes were consistent with the standard strain 17700; 16SrDNA analysis results showed that the homology rate of the strain with Vibrio alginolyticus was 99%, and it was confirmed that the strain was Vibrio alginolyticus and named Val180620. The results of the challenge showed that the Val180620 strain had strong pathogenicity to the large yellow croaker. When injected at a water temperature of 25 °C, the LD50 of the large yellow croaker of the specification (20±2) g was 2.38×106 CFU. The results of virulence-related gene analysis indicated that the strain Val180620 carried virulence-related genes such as CollagenaseVA, ToxR, FlaA and TRH, which explained that the strain Val180620 has high pathogenicity and caused ulceration and bleeding of the diseased fish. In addition, the UreR gene was not detected in the strain Val180620, but the strain Val180620 showed high pathogenicity, the result indicated that the UreR gene maybe not the determinant of the virulence of Vibrio alginolyticus..
    Isolation and identification of pathogen from juvenile pearl gentian grouper with rotten body disease

    A bacterial strain, named HW2018, was isolated from juvenile pearl gentian grouper (Epinephelus fuscoguttatus♀×E. lanceolatus♂) with skin ulcer disease and identified as Vibrio harveyi by morphological observation, physiological and biochemical characteristics and phylogenetic analysis of 16S rDNA sequences. Artificial infection experiment proved that the strain HW2018 was the causative pathogen. Death rate in one week was 100% with the injection dose (3×104 cfu/g) through celiac injection or muscle injection. The drug sensitivity test revealed that V. harveyi was sensitivity to meropenem, ciprofloxacin and levofloxacin, but resistant to ampicillin.

    Laboratory simulation experiments are carried out to explore the effect of removaling phosphorus and controlling phytoplankton from eutrophic water by using phoslock-diatomite. The result shows that the removal rate of 1.2Phl + 80 mg/L DM is the highest for TP, reaching 65.9%, followed by 1.5Phl + 80 mg/L DM, reaching 55.0%.The removal rate of SRP is mainly related to the dosage of phoslock?, however, the SRP removal rate did not change significantly when the dosage was higher than 1.2 Phl. Meanwhile, the abundance of phytoplankton decreased significantly in all experimental groups, and the phytoplankton species structure changed; 1.2Phl + 80 mg/L DM and 1.5Phl + 40 mg/L DM have a good inhibition rate, 74.1% and 81.8%. And there was no significant difference between the two groups in controling algal. In summary, using the dosage of 1.2Phl +80 mg/L DM phoslock?-diatomite can achieve better phosphorus removaling and algal controlling, In addition, it has a lower cost. So using phoslock?-diatomite is expected to be applied in the actual management of eutrophic water.

    Microalgae immobilization technology is a technology that immobilizes free microalgae cells on a carrier and changes it into a solid biocatalyst to be utilized. Immobilization can significantly improve the stability and longevity of algae cells, and is convenient for collection. At present, the "embedding method" in which microalgae cells are encapsulated in gel polymer by fixing method is often adopted due to its advantages of relatively simple operation, lower cost, less damage to cells, larger cell retention, etc. In order to explore the effect of immobilized microalgae on the removal of phosphate and ammonian from aquaculture wastewater, Scenedesmus obliquus was used as algae species, and sodium alginate embedding technology was used to carry out purification comparative tests of different algae embedding densities (S-1 group: 2.52×104cells/ml, S-50: group 1.26×106cells/ml and S-100 group: 2.52×106cells/ml), and the effects of embedding density on the growth of algae cells were also investigated. The results showed that the embedding density had a significant effect on the removal of phosphate and ammonian and the growth rate of algae cells (P<0.05). The higher the embedding density is, the better the removal effect of phosphate and ammonian is. The final removal rates of phosphate for S-1, S-50 and S-100 groups are 61.11%, 83.33% and 88.89%, respectively. And the final removal rates of ammonian are 39.95%, 50.00% and 57.53%, respectively. The main reason may be that the higher the concentration of algae cells is, the more effective algae cells participating in the reaction are and the higher the amount of nutrients absorbed is, so the efficiency of removing phosphate and ammonian per unit time is high. The growth rate K value of S-1, S-50 and S-100 groups of S. obliquus is S-1>S-50>S-100, i.e. the growth rate K value gradually decreases with the increase of embedding density of algae cells. This may be due to the fact that the larger the cell embedding density is, the greater the mass transfer resistance of CO2, O2 and nutrients is, and the lower the permeability of algae-spheres is. All these factors can cause subsequent algae cell proliferation to be affected.

    Electric stimulation of the male Cherax destructor's fifth foot base and front and rear can specifically cause contraction of the muscle near the vas deferens and can be used to obtain spermatic pods. In this paper, the voltage, frequency, duration and other conditions of electrical stimulation were optimized for the purpose of obtaining more normal sperms without damaging the shrimp body. The test results show that the stimulation voltage is 45V, the frequency is 32hz, and the stimulation time is 4s, which is the optimal stimulation condition. The spermatozoa in spermata were mature and normal, and the number of spermatozoa in a single spermatozoa ranged from 5.2×104 to 1.12×106. The pods obtained in this experiment can be used as artificial insemination materials to provide technical support for the artificial reproduction of blue devils in the sky.
    Evaluation of environmental quality and change trend in Sansha Bay mariculture area in recent ten years

    Based on the monitoring data of water quality, sediment quality and aquaculture organisms quality in Sansha Bay mariculture area from 2009 to 2018, the comprehensive state of environmental quality in Sansha Bay mariculture area was evaluated according to the comprehensive index method of environmental quality. The results showed that the comprehensive environmental quality of Sansha Bay mariculture area was with a steady and rising trend in the past decade. In 2009, the environmental quality level was only passing, and the environmental quality requirements of the functional area could not be met in some time periods. From 2010 to 2012, it was better and could generally met the environmental quality requirements. Since 2013, it has been excellent meeting the environmental quality requirements, but in 2018, the comprehensive index of environmental quality decreased slightly. This trend is related to the development of aquaculture industry, harbor industry and marine environmental protection in Sansha Bay in recent years. Through analyzing and evaluating the comprehensive situation of environmental quality in Sansha Bay mariculture area in recent ten years, the change trend of aquaculture environment is grasped, which provides basis for environmental protection of fishery waters.
    Current condition and eutrophication assessment of water quality in the main bays of Zhangzhou

    The analysis and assessment of seawater quality in the main bays of Zhangzhou was carried out, according to the results of water quality monitoring in 2017. The eutrophication degree was evaluated in the main bays of Zhangzhou. The results showed that the quality of sea water in Zhangzhou main bay was not bad on the whole. The main pollution factors were inorganic nitrogen and orthophosphates. The eutrophication of seawater was over the light, mainly occurring in dry season and peaceful season. The interannual variation and spatial distribution of nitrogen and phosphorus nutrients in seawater from 2013 to 2017 were analyzed. The corresponding countermeasures and suggestions for controlling marine pollution were put forward.
    Empirical analysis of fishery economic growth factors in Fujian Province

    In the process of implementing the 2018 rural revitalization strategy, Fujian province followed the spirit of the CPC central committee, continuously deepened the supply-side structural reform of fishery, insisted on improving the quality and efficiency, green development, and promoted the steady and healthy development of fishery economy. This paper analyzed the factors of fishery economic growth in Fujian province by using C-D production function models. The input factors included capital input, labor input, breeding area and vessel. Combined with the data and through the analysis of the econometric model, our results showed that: the output elasticity of capital was 0.1464, and the output elasticity of labor was -1.8987, which was smaller than the output elasticity of capital, indicating that the input of labor at the present stage might be an extensive increase, and the skills of practitioners were relatively weak. Modern economic theory tells us that when other conditions remain unchanged (such as the level of productivity and other input factors), the impact of blindly increasing labor force on fishery economic output is decreasing. Compared with the substantial increase of fishery economic output of capital, the amount of labor has not reached the effective allocation, which should have a certain quantity instead of a certain quality, such as whether the labor force has the corresponding fishery technology. In terms of the contribution of input factors to fishery production, firstly, the output elasticity of labor is negative, and its contribution to fishery growth is relatively limited, which also indicates that Fujian province has abundant fishery labor at present. Secondly, the contribution rate of capital to fishery growth is 7.08, which is close to 9 times that of labor. This indicates that the investment ratio of fishery capital to labor is relatively scarce, which also reflects the society's emphasis on fixed capital investment. Thirdly, although the contribution of total factors to fishery growth is negative, it means that Fujian fishery economy should mainly focus on capital input and appropriate labor input at the present stage. When the marginal economic benefits brought by capital input and labor input decline, it is necessary to consider the peripheral input factors of fishery in order to evaluate the situation. However, it does not mean that such factors, such as the implementation of fishery policy, management, fishery technology and mutual insurance, will not be input at this time, because the most important thing at this stage is matching. Therefore, the province of total factors also plays an important role in the growth of fishery economy, which cannot be ignored. The research methods of this paper are economic growth factor analysis, scale economy analysis, and then through statistical and econometric analysis methods to explore the impact of fishery economic growth factors based on the empirical analysis. Through this study, we can better improve the fishery economy growth of Fujian province to a certain extent. At the same time, it can enrich the literature on fishery economic growth. Because in the previous literature research on fishery economic growth, there are few studies on Fujian province alone.
