    Isolation and identification of Pseudomonas plecoglossicida in large yellow croaker(Pseudosciaena crocea)of Fuzhou area

    The cultured large yellow croakers(Pseudosciaena crocea)of Luoyuan bay got white nodules disease at the end of spring and the beginning of summer in 2013. The pathogen was isolated from the fishes’ spleen, liver and kidney with significant pathological signs, then its morphological, physiological and biochemical characteristics, taxonomic status, and drug sensitivity were analyzed. The 16S rDNA sequences of the pathogen was more than 99% homology with Pseudomonas plecoglossicida in P.putida group, the analysis of housekeeping gene gyrB confirmed its phylogenetic affiliation and showed identities more than 99% homology, which was also demonstrated by biochemical tests. The test result of the sensitivity to antibiotics showed that doxycycline, norfloxacin and enrofloxacin had the most inhibitive effects on the strains, while ofloxacins, levofloxacin and tetracycline had intermediate sensitive.
    Isolation and identification of a pathogen from Haliotis discus nanri

    The occurrence of disease is becoming more and more serious with the expansion of Haliotis discus nanri breeding scale, but so far no special reports focus on the research of these diseases. In order to provide some basic information for disease prevention and control of H. discus nanri, a dominant bacteria strain was isolated from the foot lesions and digestive gland of obviously dying individuals which were showing overt symptoms and proved to be the pathogen by recurrent infection experiment. The disease symptom of the pathogenic bacteria was observed carefully while biochemical characteristics and drug susceptibility test of the pathogenic bacteria were also detected separately. It turned out that the disease symptom was not significantly at the initial stage, and festering holes of different sizes could be just found on the foot of H. discus nanri after a period of time. The results of physiological and biochemical analyses showed the pathogen was Vibrio alginolyticus which was sensitive to 21 kinds of antibiotics such as flumequine and florfenicol. It highly recommend that drug sensitive experiment should be necessary to carry out every time with the purpose of screening out the appropriate effective drugs to prevent and control the V. alginolyticus in H. discus nanri production process.
    Community characteristics of phytoplankton and relationships with environmental factors in southern part area of Minjiang Estuary in Fujian Province in summer

    The present study reported on the community structure of phytoplankton and its relationship to the environment factors, based on phytoplankton water samples collected in southern part area of Minjiang Estuary from 2011 to 2014 in summer. A total of 193 species belonging to 51genera (4 phyla ) of phytoplankton in survey area were indentified. Diatom was the major phytoplankton group. The annual average cell abundance of the phytoplankton varied from 1.20×105 to 3.87×105 individual /dm3. Horizontal distribution of phytoplankton cell abundance showed a trend of decreasing gradually from inshore to offshore, inner to outer bay. But the horizontal distribution of phytoplankton biodiversity Shannon-Wiener diversity index was on the contrary of that of phytoplankton cell abundance. Statistical analysis showed that the variations of phytoplankton cell abundance were positive correlated obviously with the concentrations of silicate, phosphate, ammonia, nitrate and nitrogen. The phytoplankton growth was mainly limited by ammonia concentration, and Secondly by the nitrate concentration.
    Artificial propagation of Oreochromis niloticus by hormone combination injection

    In order to verify the practical effect on artificial propagation of Oreochromis niloticus by hormone combination injection, the spawning rate and oxytocin effect time were investigated within 144 h after hormones injection in breeding season. Meantime, morphology of the eggs squeezed from the injected females was observed. Results showed, under water temperature of 21~29℃, the spawning rate was 57.5% in the experimental group, and mostly happened within 72 h after injection, while the spawning rate was 27.5% in the control with a irregular spawning time. At 72 h after injection of hormones, 50% female in the experimental group reached maturity, in comparison with 30% in control group. These results showed that hormones combination injection could effectively improve the spawning rate, which provided some base for artificial propagation of Nile tilapia.
    Study on the development strategy of fishing harbor economic zone in China

    In this paper, the development status of fishing harbor economic zones all over China was studied. A few of problems in the development process of these fishing harbor economic zones were identified. It was found that the expanding functions of many fishing harbors were insufficient, and the industrial development was still in low level. Moreover, the equipment and technology of many fishing harbors was backward. At last, the following concrete suggestions including strengthening central guidance and overall arrangement of fishing harbor development, linking fishermen’s business restructuring together with the small town construction in fishing area, enhancing the fishing harbors’ infrastructural facilities, providing policy supports for the development of fishing harbor economic zones, and improving the opening-up level, were also proposed as responses to solve the above-mentioned problems.
    Embryonic and morphological development in larva, juvenile, and young stages of hybrid grouper (Epinephelus moara ♀×E. lanceolatus ♂)

    Hybridization is the most widely used and effective method of artificial breeding in aquaculture. We documented embryonic development and the growth and morphological characteristics of larva, juveniles, and young fish in the hybrid (Epinephelus moara ♀× E. lanceolatus ♂) by artificial methods. The embryonic development of the hybrid was divided into cleavage stage, blastula stage, gastrula stage, neurula stage, and organogenesis stage. In sea water (22±1℃, salinity 33, pH7.8), 35 h 23 min was required for hybrid embryonic development. In cleavage stage, blastula stage, before and after the larvae, the hybrid had higher mortality. Statistics showed that hybrid hatching rate was 91.5% lower than control group (93.1%), deformity rate was 10.5% higher than control group (5.3%). Based on the features of the yolk sac, second dorsal fin spine, pelvic fin spine, scales and body color, the post-embryonic development of the hybrid was divided into larval (0~27 d), juvenile(28~45 d), and young fish stages(46 d). The results suggested that the hybrid grows more rapidly than E. moara.
    Concentration distribution space-time characteristics of TPH in the saccostrea cucullata from Fujian middle southern coastal waters

    According to the investigation data of shellfish (Saccostrea cucullate) in the middle southern coastal areas of Fujian province during 2009 -2014, the concentrations and distributional feature of petroleum hydrocarbon (TPH) were studied. The result showed that the average concentration of TPH was ranked as: Weitou Bay > Quanzhou Bay >Xiamen Bay > Foutan Bay > Dongshan Bay > Dagang Bay > Jiuzhen Bay > Meizhou Bay > Shaoan Bay. The content of TPH in Weitou Bay has exceeded the standards seriously. The concentration variation of TPH was undulating in each monitoring coastal areas, i. e. being strong in the year of 2010 and 2013. The pollution index of TPH ranged from 0.08 to 23.3. About 54 samples were tested in six years, the severe pollution of which accounted for 26 %,whereas mild case accounted for 37 %, and with the 37 % of the samples were no detected.
    Simultaneous determination of PFOS and PFOA in sediments samples using liquid chromatography-mass spectrometry

    A method for simultaneous determination of PFOS and PFOA in environmental sediments samples was developed by ultra pressure liquid chromatography coupled with tandem mass spectrometry. Sediments sample was ultrasonic extraction with 15 mL methanol twice, then enriched and purified through a C18 SPE, and finally concentrated into 1.0 mL with an automated de-solvent device. Target analytes were separated on a Eclipse Plus-C18 column using methanol (containing 5.0 mmol/L NH4COOH) –water (containing 5.0 mmol/L NH4COOH) as mobile phase. The negative electrospray ionization (ESI-) and multiple reaction monitoring mode (MRM) was utilized. The recoveries (n=3) and relative standard deviations(RSD,n=7) were 82.5± 9.2% and 8.3% for PFOS at 0.5 ng/g, 2.0 ng/g and 5.0 ng/g spiking levels and 103.2± 9.2% and 9.1%for PFOA at 0.5 ng/g, 2.0 ng/g and 5.0 ng/g spiking levels,respectively. An internal standard calibration method was chosen to the quantitative analysis. The calibration curves of PFCs were linear in the range of 0.5-20 ng/g with correlation coefficients more than 0.999 0.The detection limits were 0.08 ng/g for PFOS and PFOA. The results showed that this method possessed efficiency and good selectivity,and can be used for the detection of PFOS and PFOA in marine sediments.

    Nesfatin - 1 is the hypothalamus gene encoding nuclear protein - 2 (nucleobindin - 2, NUCB2) produces a neuropeptide after cracking. Studies have shown that nesfatin - 1 play an important role in reproductive regulation, in order to study the Nesfatin - 1 relation with Epinephelus coioides reproductive regulation, in this paper, we have cloned two NUCB2 (NUCB2a and NUCB2b) gene, analyzes NUCB2 coding protein get sequence of amino acids about two nesfatin - 1 (nesfatin - 1a and nesfatin– 1b), at the same time, we analysis of the two kinds of nesfatin– 1's signal peptide and mature peptide, found it is consistent with that of vertebrates. Through real-time quantitative PCR technology, we determined the mRNA expression level of Epinepheluscoioidesnesfatin-1 in different tissues. The expression patterns revealed that, nesfatin-1a and nesfatin-1b widely expressed in the brain tissue, especially nesfatin - 1b abundantly expressed in the pituitary, also has the corresponding expression in the gonads. We determined thenesfatin-1 mRNA expression level of hypothalamus during ovary development, the result reveal that, nesfatin - 1a and nesfatin– 1b in the ovaries Ⅵ period (degradation of yolk formation stage oocytes) significantly increased, showed that two kinds of nesfatin - 1 are likely to participate in the regulation of ovarian mature. The intraperitoneal injection experiment results show that, injecting nesfatin - 1a and nesfatin – 1b 15 min, both GnRH1 and GnRH3 mRNA expression level of the hypothalamus are reduce, but in the pituitary LH –βand FSH – βmRNA expression level has not changed. Injection of nesfatin - 1a and nesfatin – 1b 60 min, the mRNA expression level of the hypothalamus GnRH1 and GnRH3 still suppressed. Injected nesfatin - 1a and nesfatin – 1b after 60 min, there have no effect change on the expression level of LH - β, but significantly reduced the expression of FSH -β. Through the above study shows nesfatin - 1a and nesfatin– 1b involved in Epinephelus coioides reproductive regulation.

    Known as the major marine shellfish aquaculture country in the world, China has paid much more attentions on the marine biological industry, public health and nutrition requirements. High value utilizations of marine shellfish and their byproducts is the mainstream focus point of the marine development and management. Glycosaminoglycan is an important marine shellfish substance that has many bioactive properties, including anti-clotting, hypolipidemic, anti-tumor, anti-oxidation, strengthening immune system, etc. The current stage of knowledge on the marine shellfish glycosaminoglycan is reviewed in various aspects: extraction, isolation, purification, characterization and biological activities, which lays the foundation for the fine processing and comprehensive utilization of shellfish and its cooking liquor, as well as providing scientific basis for the developments of marine shellfish resources utilization and marine health care products.
    Deepening fishery academic exchanges and promoting the development of fishery cooperation between Fujian and Taiwan

    Fujian and Taiwan are close neighbors across the Taiwan Strait, and the fisheries exchange and cooperation has a long history between two provinces. Based on years of work experience of engaging in fisheries academic exchanges between Fujian and Taiwan, the author reviewed the main situation and achievement, and proposed several new measures such as strengthening fisheries academic exchanges platform building between Fujian and Taiwan, expanding platform’s exchange and cooperation functions, playing vital roles of professional committee and members, and enhancing fisheries academic information exchanges and cooperation between Fujian and Taiwan. Therefore, the cross-strait fishery science and technology exchanges and cooperation could be promoted to a higher level.
