    Molecular characterization of six strains of Aeromonas hydrophila isolated from fish as well as detection and analysis of virulence genes

    Six strains of Aeromonas hydrophila isolated from dying fish were molecularly characterized as well as five virulence genes were detected and analyzed in order to evaluate the feasibility of detecting the virulence genes to infer the potential pathogenicity of A. hydrophila at the molecular lever. 16S rDNA and gyrB gene were used to characterize A.hydrophila, five virulence genes including aerolysin (aer), haemolysin (hly), serine protease (ahp), heat-stable cytotonic enterotoxin (ast) and heat-labile cytotonic enterotoxin (alt) were detected by PCR, and the sequences of gene and virulence were analyzed using Mega 5.2. All of the strains were characterized as A.hydrophila, and four of five virulence genes at least were detected as positive, including the genes of haemolysin and two kinds of cytotonic enterotoxin. By the sequences analysis, we found the amino acid sequences of high similarity among aerolysin, haemolysin and serine protease. A.hydrophila could be well characterized using 16S rDNA and gyrB gene sequences, and the varied virulence genes indicated the potential pathogenicity. Through the sequences analysis, the genes of aer, hly and ahp were relatively conservative, and the encoding virulences were highly conserved. Detecting the virulence genes of aer, hly and ahp could infer the potential pathogenicity of A. hydrophila in the clinical molecular diagnosis.
    Preliminary study on the physiological disorders of Porphyra haitanensis from Fuding Xiamen Bay

    The rotten disease of Porphyra haitanensis from Fuding Xiamen Bay was investigated in this paper,the samples was collected and took back to culture in laboratory, and then the occurrence and development process of physiological disorder were studied by using microscopy methods. The main symptoms of this disease were faded, pale and then completely rotten in tip part of thallus. No obvious lesions rotten hole was found. Main pathologic symptoms included, in tip part of thallus, cell edge blurred, pigment body color changed and then developed into cell edge disorder, cytolysis, protoplasm outflow, abnormal cellular structure.The severe lesions could easily cause secondary infection by bacteria and protozoa, resulting in ulceration of the thallus, and finally caused the thallus dissociated completely. Rainy weather could easily lead to this disease to break out. Cutting rotten parts of the thallus and extending the drying time in sunny day were the key measures to control this disease.
    Analysis of volatile compounds of fresh and liquid-smoked Pseudosciaena crocea by Chromatography - Mass Spectrometry

    The headspace solid-phase microextraction-gas chromatography-mass spectrometry method was used to analysis volatile compounds of the fresh and liquid smoked Pseudosciaena crocea. Totally 58 and 81 volatile components were detected in fresh and liquid smoked Pseudosciaena crocea. The flavor substance mainly contained hydro carbons and ester compounds before liquid smoked treatment, respectively 16 and 10; mainly contained phenols and ketone compounds after liquid smoked treatment , respectively 20 and 14. The results showed that the liquid smoked Pseudosciaena crocea were changed largely in flavor substance, a marked increase in flavor compounds.
    Determination of inorganic arsenic in seaweeds with HPLC-HG-AFS

    This paper is about the research of the determination of inorganic arsenic in edible seaweeds with High performance liquid chromatography-hydride generation-atomic fluorescence spectrometry(HPLC-HG-AFS). The curved surface fitting method was applied to establish optimum conditions of extraction reagent, time and temperature based on the inorganic arsenic contents of Sargassum fusiforme. It was found that the optimum conditions is with 0.1%HCL at 52.5℃ for 76min. The detection limit of methodology was 0.0635mg/kg. The recovery rates of 1.0 mg/kg and 10.0mg/kg in samples were between 80.5% and 105.7% and the relative standard deviations were between 4.5% and 5.0%. The content of inorganic arsenic of sixty types of laver from different areas in Fujian province were examined. The results showed that inorganic arsenic concentration in laver was below,which accounted for the total arsenic concentration of 0~1.96%.
    Investigation and protection on fish resource of Chongyang river in Jianyang City

    The composition and distribution of fish was investigated at 4 sections sampling in Chongyang river during October of 2007 to February of 2012. The results showed that a total of 75 fish species were collected, belonging to 5 orders, 16 families, 54 genera. Cypriniformes constitutes the dominant component of species composition, and is consist of 4 families, 39 genera and 51 species, accounting for 68% of the total species,Siluriformes rank is the second including 4 families, 6 genera and 11 species, accounting for 14.7% of the total species. According to the family of statistics,Cyprinidae consists of 40 species ana is the most common species, accounting for 53.3% of the total species, Bagridae is the second common species, consisting of 7 species, accounted for 9.3% of the total species. The species composition collected from 4 sections is almost the same,but the main species composition that the ratio of single species ot Iish to the total catch in weight in this river is greater than 5% are different obviously. The average daily catch of fish for single ship or bamboo rafts in 4 sections is different,the highest is Huiyao section,and the lowest is Lushang section. Individual fish caught in size and age tends to be miniaturization and young.
    Horizontal distribution characteristics and influencing iactors of heterotrophic bacterial abundance in Dongshan Bay in spring and summer

    Based on two cruises which dated from May 2011 to August 2011, the horizontal distribution patterns of heterotrophic bacteria in Dongshan Gulf were studied. Moreover, the relationship between the bacterial abundance and the environmental ecological factors was also analyzed. The results showed that the mean bacterial abundance in Dongshan Gulf in spring and summer were 2.05×103 mL-1 and 2.82×103mL-1 respectively. The bacteria abundance of surface water in Dongshan Gulf was higher than that of the bottom water in spring, while the bacteria abundance of surface and bottom water was well distributed in summer. Besides, the bacterial abundance in the Gracilaria lemaneiformis culture area was highest in spring in Dongshan Gulf while that in the upwelling area and fish-cage area was higer in summer. The correlation analysis showed that the bacterial abundance closely correlated with the concentration of reactive silicate, chlorophyll-a, dissolved oxygen (DO) and COD in spring and by contraries in summer. Meanwhile there was no significant correlation between the bottom of the bacteria abundance and the environmental ecological factors. Based on the standard of sea-water, it showed that Dongshan Gulf was less affected by terrestrial pollution using the number of heterotrophic bacteria as evaluating indicator.
    Survey and assessment of seawater quality and nutritional status at north sea area of Dongdai village in Nan Ri island in summer, winter and spring

    Abstract According to the survey of seawater quality and nutritional status at sea area around Dongdai village in Nan Ri island in June, October in 2012 and March in 2013, the result suggested that the quality of seawater in this area is good, inorganic nitrogen, chemical oxygen demand and oil are in accordance with Chinese national 1st class water quality standards. On the contrary, the inorganic phosphorus reached the 3rd class of Chinase national water quality standards in February voyage in 2013. The phosphorus-rich phenomenon is obvious. By the value of E, TSIm and WQI, the seawater in February voyage in 2013 come out to be light pollution, and the key factor leading to it is inorganic phosphorus. It could be ascribe to the abalone nursery industry. Overall, seawater in this area is in a healthy state, and can meet the natural P. haitanensis growth requirements, but the phosphorus-rich phenomenon should be attention to, without which could lead to environmental degradation.
    Determination of phthalate esters residue in aquatic products by solid phase extraction with gas chromatography-mass spectrometry

    A method was developed for the determination of 6 common phthalate esters(PAEs) in aquatic products by gas chromatography-mass spectrometry(GC-MS). The PAEs were extracted by cyclohexane-ethyl acetate(1:1) mixed solution, degreased by acetonitrile saturated n-hexane, then extracted by N-hexane saturation of acetonitrile, and purified by PAE30006-C SPE glass column, washed by 1% ethyl acetate and eluted by hexane-ethyl acetate(1:1), the eluent was concentrated and dissolved in hexane, and analyzed by GC-MS. The results were showed that: the linear range of 6 PAEs were 10~5000 ng/mL, and the correlation coefficients were 0.9992~0.9998, limits of detection(LOD) were 0.62~2.96 μg/kg, limits of quantifications(LOQ )were 2.0~9.0 μg/kg, The average recovery of the added test in tilapia from 10,50,100 μg/kg was 70.5%~105.4%,and the relative standard deviations(RSD) was 3.78%~7.57%. The average recovery of the added test with 50 ug/kg in Pacific white shrimp, grass carp, mud crab, yellow croaker and oyster was 71.5%~102.5%, and the RSD was 3.25%~7.86%. The method was suitable for the determination of PAEs in aquatic products with high extraction efficiency and purification effect.

    The pesticide residues in environment water could not be detected directly, because the content was much lower than the limit of detection (LOD) of analysis instrument. The bottle-neck was how to enrich the targets. In this paper, it was reviewed that the research progress of enrichment and purified technology of the pesticide residues in environment water in recent years. It was focused on the discussion of influence factors of liquid - liquid extraction (LLE), such as: the selection of extraction solvent, mode of vibration, mode of condense, phenomenon of emulsification and salt out, pH, purification of extractions; of solid phase extraction (SPE), such as: the selection and activation of absorbent material, breakthrough volume, sample flow rate, moisture removal, leaching of the SPE cartridge, elution of the targets, pH and NaCl, purification of the elution solvent. Some new type enrichment methods were briefly introduced, and their disadvantages were discussed. The directions for future research were forecasted on the enrichment and purified technologies of pesticide residues in environment water.

    In order to improve the hatching rate and yield,the dried Cyclina sinensis were hung on the bamboo raft, then stimulated by flowing water and induced spawning 63 times in 34 earth ponds of a total area of 100.17 hm2 from 2007 to 2011. 815.055 billions of fertilized eggs and 197.074 billions of D-shaped larvae were obtained. The hatching rate was 24.18% . The larvae metamorphosed into 42.385 billion juveniles, the metamorphosis rate was 21.51%, the seedlings of 0. 5 cm shell length was 37.702 billion, the rearing rate was 88.95% , harvested average 376 million seedlings per hectare, it achieved the requirements of large—scale yield breeding.
