    Investigation community structure of Phytoplankton in Yueqing Bay

    Through the investigation in May and September, the Phytoplankton species composition,quantity distribution,diversity index in Yueqing Bay was researched The results showed that there were 5 phylum 37 genuses 62 species in May and there were 5 phylum 43 genuses74 species in September. The Phytoplankton were composed of Diatom and Dinoflagellate. Diatom was absolute predominance judged from species and abundances. The Dominant species of May were Skeletonema costatume and Coscinodiscus jonesianus, and the Dominant species of September were Skeletonema costatume and Planktoniella blanda. The Cells number of phytoplankton assemblage of Septmber were more than May of 16 sites, but the distribution quite of May was even than September. the Shannon-Weiner diversity index、Margalef richness index and Simpson dominance index of September were more than the May, but the Pielou evenness index was less than than the May. Through the study of the Zooplankton species composition、quantity distribution、diversity and relationship between abundance and environmental factors in Yueqing Bay is to provide evidence for ecological monitoring, evaluation, and provide scientific evidence for the development of the sustainable utilization of marine resources to the sea.
    Study on the ecological characteristics of phytoplankton community in Minjiang Estuary

    Investigations on phytoplankton community and environmental factors were carried out in Minjiang Estuary of Fujian , in May (spring) and November (autumn) of 2009 . The total 129 species in 54 genera were found out in this sea area . Taxonomically , 118 species in 46 genera were Bacillariophyta , accounting for 91% of all species ; 8 species in 5 genera were Pyrrophyta , accounting for 6% ; 1 species in 1 genus was Cyanophyta , accounting for 1% ; 1 species in 1 genus was Chlorophyta , accounting for 1% ; 1 species in 1 genus was Cryptophyta , accounting for 1% . The dominant species in spring were Skeletonema costatum , Chaetoceros curvisetus and Chaetoceros debilis ; and the dominant species in autumn were Skeletonema costatum , Coscinodiscus subtilis and Coscinodiscus jonesianus . The cell abundance of phytoplankton ranged from 1.56×103 to 7.43×105cell/L . The abundance of phytoplankton was significantly correlative to the water temperature and chlorophyll-a concentration in spring . However , There were significant negative correlation of phytoplankton abundance to salinity,and positive corelations of phytoplankton abundance to DIN concentration in autumn .

    Abstract:Each month from February 2008 to February 2009,the number of fecal coliform bacteria (FCB) in the two major Crassostrea gigas culture area (Xiagong and Jitou) of Luoyuan Bay and its seawater environment were carried out an annual monitoring survey. The survey results show that: the number of FCB in seawater between <20 to 2400 / L, with an average of 400 / L; the number of FCB in Crassostrea gigas between 2300 to 240,000 / kg, with an average 25,030 / kg. The culture area seawater compliance rate was 92.3%, the culture area Crassostrea gigas compliance rate was 26.1%. Seasonal variation: the number of FCB in seawater is summer> spring> autumn> winter; the number of FCB in Crassostrea gigas is autumn> summer> winter> spring. The FCB pollution is more serious in the two major Crassostrea gigas culture area (Xiagong and Jitou) of Luoyuan Bay .Should be strengthened shellfish products, especially eaten raw shellfish safety listing and promote the governance of land-based sources of pollution
    Determination of Bacitracin residues in farming waters by HPLC-MS/MS

    Set up a method detecte BTC residues in farming waters by HPLC-MS/MS . On the basis of analysis the extraction, purification BTC conditions, the study compared the effect of different solid-phase extraction columns, and optimized the steps of solid-phase extraction column. The linear range between 5 ng/mL~5000 ng/mL, the detection limit is 5 ng/mL, the quantification limit is 10 ng/mL. Select sea water of 3 different places to validate the method, the average recovery ranged from 79.1%~99.0% and the RSD between 3.56% ~6.69%.
    Applications of standard addition test and quality control chart in drug residues detection of primary aquatic products

    The quality control methods of standard addition test and quality control charts were introduced. According to the standard addition detection of malachite green and nitrofuran metabolites in primary aquatic products by LC/MS, the standard addition recoveries were analyzed. The quality control charts were drew by Origin, and the correlation of standard addition recoveries was analyzed by SPSS. The results indicated that the quality control charts could monitor the test results effectively, and they could ensure the detection results were accurate. The main reason which caused the variation of standard addition recycles was random error. The recoveries correlation between color malachite green and leuco malachite green was weak. The average standard addition recycles of leuco malachite green was higher than that of color malachite green. In four kinds of nitrofuran metabolites, the recoveries correlation between AOZ and AMOZ or SEM was better than others. The average standard addition recycles of AOZ was maximum, and the average standard addition recycles of SEM was minimum.

    Used Schall oven method to accelerate oxidation for researching the antioxidant effect on squid liver oil by Tea polyphenols , Natural tocopherol and Ascorbyl palmitate.The best blend antioxidant was confirmed by surface response, and the antioxidant effect was a contrast to single antioxidant. The result showed that blend antioxidant were composed of Tea polyphenols 0.28‰, Natural tocopherol 0.28‰, Ascorbyl palmitate0.14‰, the peroxide of squid liver oil was 7.93 meq/kg; and Shelf life of squid liver oil was extended from 48d to 96d which illustrate blend antioxidant has good antioxidant effect.
    Optimization of Extraction Technology of Polysaccharides from Grateloupia filicina

    Using the yield of crude polysaccharides from grateloupia filicin as index, this thesis compared and analysed several extraction methods, including water extraction, alkali extraction, enzymatic hydrolysis and microwave-assisted extraction. Using the yield of polysaccharides from grateloupia filicin as index, this thesis studied the factors that influence the index, including treatment temperature, time and S/L ratio,through the single-factor experiment and the orthogonal test, the optiamal cripness of extraction technology of polysaccharides was established. The results indicated that water extraction was the best method to extract polysaccharides and the extraction efficiency was 55.64%t, the optimal extraction technology conditions were determined to temperature of 100℃,time of 5h,solid-liquid ratio of 1:60,the yield of polysaccharides up to 59.42% .
    Analyze the bacterial detection of ruditapes philippinarum stored in different conditions

    The survival rate, colony count, the number of Escherichia coli and the number of vibrio of ruditapes philippinarum stored at 15 ℃, 4 ℃ and 18 ℃ changing with time were determined, and the correlation between them were analyzed in this paper. The results showed that, the survival rate of ruditapes philippinarum at 4 ℃ at the time of 96h was also 100%. The colony count, the number of Escherichia coli and the number of vibrio of ruditapes philippinarum at -18 ℃ showed no significant change according to the increase of storage time, were lower than the values at 0h respectively. In contrast, the colony count and the number of vibrio of ruditapes philippinarum at 15 ℃ and 4 ℃ showed the rising trend of different levels, meanwhile, the correlation coefficient between them at 15 ℃ and 4 ℃ were significant, respectively 0.884, 0.927. However, the number of Escherichia coli showed no significant change at 15 ℃ and 4 ℃.
    The different influence of soft pellet and fresh trash fish surimi to Pagrosomus major infected by Cryptocryon irritans

    Abstract: We divide 6 indoor cement pools into 2 groups in our study (treatment group I, control group II), each group has 3 parallels. There are 3000 Pagrosomus major in total, each indoor cement pool has 500 Pagrosomus major (initial average weight 70.0 g). Soft pellet and fresh trash fish surimi were respectively added into treatment group and control group to explore the effect of the infection rate of Cryptocryon irritans to Pagrosomus major. The results show that Pagrosomus major were be infected by Cryptocryon irritans after the addition of both soft pellet and fresh trash fish surimi, however the infection rate were showed significant difference (P<0.05). Fresh trash fish surimi feed will much more easier lead to a spread and outbreak of Cryptocryon irritans infection than soft pellet. The major infected areas are fish gill filaments and body surface.
    Preliminary investigation on the resource of Coelomactra antiquata in Changle coastal waters

    The resource of Coelomactra antiquata was investigated in Changle coastal waters in October 2012. 9 samples were collected at only 4 investigation sites, accounting for 12.9% of the all sites. The shell length of the samples is 27.11mm~113.55mm, average of 61.29mm; the body weight is 2.0g~162.0g, average of 41.8g. The maximum value of distribution density and biomass of Coelomactra antiquata was 144.30 ind./hm2 and 4.98kg/hm2 respectively. The results show that the resource of Coelomactra antiquate in Changle coastal waters was declined obviously.

    Many people died for eating nocuous nassarius or puffer fishes which contain neurotoxic tetrodotoxin in southern China.The paper analyzed the application of the collaurum chromatography rapid detection card in nassarius and puffer fishes, and confirmed detection limit of kits was about 0.5MU/mL or 100ng/mL in tetrodotoxin aqueous solution. On the basis of limit value of tetrodotoxin, the rapid detection kits were designed for screening whether toxic content was above limit standard(10MU/g) in 20 samples including nassarius and puffer fishes, and precision rate reached 100%, single sample testing time was about 20 minutes. The kits have many advantages such as high sensitivity, good specificity, user-friendly control, rapid identification, non- apparatus supplementary, unaided eye results reading, and etc. The testing kits can be applied to testing organization at grassroots level, fishery management grassroots units, and aquatic product processor for screening tetrodotoxin in fishes and snails.

    By the introduction and breeding test of new Undaria pinnatifida variety in 2010, the appropriate environmental conditions and breeding technologies of farming Undaria pinnatifida in southern China has been explored. Technologies such as key points of daily management and optimum water layer of breeding Undaria pinnatifida had been summed up for farming in Guanwu Sea area, Fujian province, Lianjiang County, Xiaocheng townships. The problem of had seedlings cropped by Siganus fuscescens had also been resolved. By compare the outcome of Undaria pinnatifida and sea tangle breeding, the conclusion had been drawn that Undaria pinnatifida breeding surpasses sea tangle as long as the benefit is concerned. All these build a solid foundation for the extension of Undaria pinnatifida breeding in Fujian province or in southern China.
