一株斜带石斑鱼(Epinephelus coioides)病原菌的分离与鉴定

Isolation and identification of a bacterial pathogen from Epinephelus coioides

摘要: 2 0 1 1 年7 月,福建省晋江某养殖场养殖的斜带石斑鱼(Epinephelus coioides、发病,病鱼体重3 ~ 1 0 g ,症状为鱼体色变深,拒食, 离群缓慢游动,背鳍下方局部鱗片脱落、表皮溃烂,肝脏点状发红。2 0 1 1 年7 月从濒死病鱼肝脏、肾脏中分离到2株細菌,分别命名为201107 - 1 与201107 -1 1 ,经生理、生化指标和16S rR N A 基因鉴定,这两株均为哈维氏狐菌(V ibrio harveyi ) 。毒性实验发现这两株菌都能引起斜带石斑鱼表皮溃烂症,其中201107 - 1 1 毒性较强,被用于后续感染实验。通过肌肉注射、腹腔注射与创伤浸浴3 种方式进行回归感染实验,得到3 种方式对石斑鱼的半致死量LD5。分别为4 .3 × 1 0 4、2. 9 × 105和5. 4 ×106 c fu /g 鱼体重。从人工感染的石斑鱼肾脏中再次分离到该菌,经生理、生化指标和16S rR N A 基因鉴定,为同株菌。药敏实验结果表明,该菌株对红霉素、大观霉素、氯霉素等1 6 种抗生素高度敏感,对吱喃唑酮、克拉霉素、环丙沙星等9 种抗生素中度敏感,对苯唑西林、青霉素G、万古霉素等1 3 种抗生素不敏感。本研究对石斑鱼病害防治与健康养殖具有一定的指导意义。

Abstract: Serious disease outbreak occurred in a groupers Epinephelus coioides culture farm in Jinjiang,Fujian province in Julv 2011. ih e weight oi diseased groupers was 3 ?10 g. The characteristics oi aiseased grouper were observed,displaying body colour darkening,feeding refused, swimming slowly,and redness of the liver and spleen spot. Two strains of bacteria were isolated from the liver and spleen of moribund groupers and designated as strain 201107 - I and 2011107 - II in July 2011. A ll the two isolates were identified as Vibrio harveyi by physiological and biochemical tests combining with 16S rRNA gene sequence analysis. A rtificial in jection demonstrated that strain 201107 - 1 and 201107 - II were both pathogenic to E. coioides . Strain 201107- II showed the stronger toxicity to E. coioides and was used to the subsequent analysis. Three experimentaltypes of artificial infections were conducted, muscle injection, celiac injection and wound immersion, to confirm the toxicity of strain 201107- II ,and the half lethal dose ( LD5。) to E. coioides was 4. 3 × 104 , 2. 9 ×105 and 5. 4 × 106 c fu /g fish weight,respectively. We also isolated a bacterial strain from this artificial infected E. coioides,and this strain was identified as the same strain as the 201107 - II using traditional physiological and biochemical characterization along with molecular 16S rRNA gene analysis. The sensitivity tests of antibacterial agents indicated that the bacterial strain 201107 - II was sensitive to erythromycin,spectinomycin,chloramphenicol,and other 13 agents ; middling sensitive to furazolidone, clarithrom ycin, and ciprofloxacin and other 6 agents; resistant to oxacillin,penicillin G,vancomycin and other 10 agents. This study will provide necessary data about the sustainable and healthy development of E . coioides culture
