Box-Behnken响应面法优化孔石莼多糖络合锌工艺

摘要: 本研究以孔石莼多糖锌络合物的锌含量为指标,采用3因素3水平的Box-Behnken响应面法优化孔石莼多糖络合锌工艺,探究了反应温度、反应体系pH值、孔石莼多糖与Zn2+质量比对孔石莼多糖络合锌反应的影响。结果表明,孔石莼多糖与Zn2+的最佳络合条件组合为反应温度50 ℃、反应体系pH 5.0、孔石莼多糖与Zn2+质量比为2.8∶1,在此条件下,控制反应体系Zn2+浓度为3.00 mg/mL并进行反应90 min,络合效果最佳,孔石莼多糖锌络合物锌含量为(102.41±0.67)mg/g。

Abstract: In the present study, the Box-Behnken design and the method of response surface (RSM) analysis with 3 factors and 3 levels was employed to optimize the complexation conditions between Ulva pertusa polysaccharide and Zn2+, and investigate the effects of temperature, pH and weight ratio between Ulva pertusa polysaccharide and Zn2+ on the contents of zinc in polysaccharide-Zn. The results showed that the optimum conditions of Ulva pertusa polysaccharide complexation Zn2+ required temperature as 50 ℃, pH as 5.0, polysaccharide-Zn2+ weight ratio as 2.8:1, the concentration of Zn2+ as 3.00 mg/ mL, and reaction time as 90 min. Under this condition, the contents of zinc in polysaccharide-Zn reached the maximum value as (102.41 ± 0.67) mg/g.
