    Optimization of decalcification technique of fish scale by response surface methodology

    • Response surface analysis was applied to optimize the decalcification technique of Tilapia mossambica fish scale. Hydrochloric acid concentration, substrate concentration, reaction temperature and reaction time were chosen as the impact factors based on decalcification rate of fish scale, and the appropriate levels and factors were selected in previous single factor experiments. According to the Box-Behnken central composite design, in accordance with the dynamic equation model, the significance of all factors and interactions among them were analyzed. The optimum condition of decalcification technique of Tilapia mossambica fish scale was obtained as follows:hydrochloric acid concentration of 2.9%, substrate concentration of 6.3%, reaction time of 1.9 h, and the decalcification rate of fish scale could reach 99.55% under this condition.
