环丙沙星单克隆抗体的制备及其特性分析

摘要: 以人工合成的牛血清白蛋白环丙沙星抗原(CPFX-BSA)免疫Balb/c小鼠,采用杂交瘤技术制备抗CPFX-BSA的M cAb,获得1株稳定分泌针对CPFX药物的单克隆抗体细胞株5G4,腹水效价达1.28×10-6;SDS-PAGE电泳分析CPFX M cAb 5G4蛋白重链和轻链的分子量约为50KD和25KD;抗体亚型为IgG3。根据ic-ELISA标准工作曲线,IC50值为192.44ng/mL,最低检测限为16.29ng/mL,与恩诺沙星交叉反应率为62.45%。制备的环丙沙星单克隆抗体为试剂盒的研制、组装奠定了基础。Abstract: The artificial ciprofloxacin-bovine serum albumin (CPFX-BSA) was inactivated as antigen to immunizate Balb/c mice. with hybridoma technology, One strain hybridomas secreting McAb against CPFX-BSA named CPFX 5G4 was obtained ultimately. The results showed that the titer of CPFX 5G4 was 1.28×10-6; the molecules of the Heavy chain and Light chain of the McAb were approximately 50 KD and 25 KD according to SDS-PAGE electrophoresis and the antibodies subtypes was IgG3. A ic-ELISA calibration curve was drawn up according to established system. It was calculated that the IC50 value was 192.44 ng/mL and the detection limit was 16.29 ng/mL. The rate of cross reaction between McAb-CPFX and enrofloxacin was 62.45%. The McAb-CPFX laid a foundation for the study and assembly of the rapid test kit.
