电刺激获得观赏螯虾天空蓝魔精荚条件的优化

    Optimization of the conditions for obtaining spermatophore of yabby by electrical stimulation

    • 摘要: 电刺激天空蓝魔(Cherax destructor)成熟雄性的第五步足基部,能够较专一地引起输精管附近肌肉的收缩,以此获得精荚。本文在不损伤虾体的情况下,以获取更多正常精荚为目的,对电刺激的电压、频率和刺激时间等条件进行优化。试验结果显示:刺激电压为45 V、频率为32 Hz、持续刺激时间为4 s,为最优刺激条件,这时能够达到75%的精荚排出率,且精荚及精荚内精细胞形态正常。精荚内精子数量在5.2×104~1.13×106之间。本次试验获取的精荚可作为人工授精的材料,为天空蓝魔的人工繁殖提供技术支撑。

       

      Abstract: Electrical stimulation can induce muscular contraction.The spermatophores of Cherax destructor were ejected by electrical stimulation around the coxa of the fifth paraeiopod.In order to obtain more spermatophores,the conditions(voltage,frequency and duration) of electrical stimulation,without injury to crayfish,were optimized step by step.The results showed that the optimum conditions were voltage as 45 V,frequency as 32 Hz and duration as 4 s,which could get 75% spermatophore extrusion rate.The cellular morphology of spermatozoa in the spermatophore was normal.There were 5.2×104 to 1.13×106 spermatozoa in each spermatophore. This paper was important for providing technical support to artificial propagation of C.destructor.
