富含ω-3多不饱和脂肪酸的磷脂酰丝氨酸制备工艺研究

摘要: 本研究以南极磷虾油、L-丝氨酸为原料,在磷脂酶D(Phospholipase D-catalyzed,PLD)的催化作用下发生转酰基反应,制得富含二十碳五烯酸(Eicosapentaenoic acid,EPA)、二十二碳六烯酸(Decosahexaenoic acid,DHA)的磷脂酰丝氨酸(Phosphatidylserine,PS)。通过单因素和正交试验,确定了PS的最佳制备工艺条件:以乙酸乙酯为有机溶剂、水相pH 4.5、有机相与水相体积比1∶1、反应温度40℃、加酶量55U、L-丝氨酸与南极磷虾油质量比16∶1、反应时间8 h,在此条件下,PS得率为85.62%,产物中EPA含量为24.33%,DHA含量为19.95%。Abstract: Phospholipase D-catalyzed transphosphatidylation from phosphatidylcholine of Antarctic krill oil to produceω-3 polyunsaturated fatty acid-riched phosphatidylserine in this study.Single factor and orthogonal experiments showed the optimal preparation conditions were:using ethyl acetate as organic solvent,aqueous phase pH 4.5,volume ratio of organic phase to aqueous phase 1∶1,reaction temperature 40℃,enzyme amount 55 U,mass ratio of L-serine to krill oil was 16∶1,reaction time 8 hours,the yield of phosphatidylserine was 85.62%,EPA content was 24.33%,DHA content was 19.95%.
