高效液相色谱在鉴定赤潮优势藻中的应用

Application of high performance liquid chromatography in determination of the preponderant harmful algae species

摘要: 本文对福建沿海赤潮优势藻进行了分离鉴定及纯种培养,运用高效液相色谱(HPLC)技术分析了赤潮藻的光合色素,并将色素组成比进行聚类分析。然后将聚类分析结果与藻类形态和分子鉴定结果比对。结果表明,在传统分类方法基础上,结合光合色素的生化标志物的特性进行进一步判断,可以实现对不同藻类更加准确的分类,能够弥补传统分类学上遇到的因种类形态差别细微而不易辨别及容易主观误判的不足,可以作为藻类形态学和分子方法分类的有效补充。

Abstract: Algae blooms is one of the great ecological disasters in China, which greatly damages to human health and sustainable development of economy. In this study, we isolated and identified 10 species of the preponderant harmful algae and 2 species green algae in the coastal area of Fujian Province. Then detected the absorption spectrum of photosynthetic pigments (ASP) in the pure cultures by high-performance liquid chromatography (HPLC) technology and conducted cluster analysis. Subsequently, we compared the ASP results of pure cultures with traditional classification. The study indicated that the further identification based on ASP could achieve more accurate classification of different algae, which could be make up the shortcomings of the traditional classification for confronting with the subtle differences of morphology. And it could be an effective supplement to algae morphology classification and molecular-biology taxonomic.
