盐酸土霉素在奥尼罗非鱼体内的药代动力学及残留研究

Pharmacokinetics and residues of Oxytetracycline hydrochloride in hybrid tilapia(Oreochromis aureus×O.niloticus)

摘要: 在水温(25±2)℃条件下,以15 mg/kg鱼体重的剂量给奥尼罗非鱼单次口灌盐酸土霉素,采用高效液相色谱法测定血浆和肌肉组织中的药物浓度,研究盐酸土霉素在奥尼罗非鱼体内的代谢及消除规律。结果显示:血药时间数据符合一级吸收二室开放模型,半衰期(T1/2Ka、T1/2α、T1/2β)分别为4.79 h、4.10 h、45.20 h,最大血药浓度为1.50 μg/mL,达峰时间为7.30 h,药时曲线下面积(AUC)为42.35 μg?h/mL。肌肉作为可食性组织,选取肌肉组织作为残留检测的靶组织,以0.1 mg/kg为最高残留限量,在本试验条件下,建议休药期不低于10 d。

Abstract: In order to study the metabolism and elimination of Oxytetracycline hydrochloride (OTC) in hybrid tilapia (Oreochromis aureus×O.niloticus), single oral administration at dose of 15 mg/kg of OTC has been investigated in healthy hybrid tilapias at (25±2) ℃. The result demonstrated that plasma concentration-time datas of OTC were best fitted using a two-compartmental open model. Absorption, distribution and elimination half-life were4.79,4.10 and 45.20h, respectively. The maximal plasma concentration was 1.50 μg/mL, peaking at 7.30h. Area under the plasma drug concentration-time curve was 42.35 μg?h/mL. As an edible tissue, muscle was selected as target tissue in this experiment. The maximum residue limit (MRL) is 0.1 mg/kg in muscle, the withdrawal time should not be less than 10 days under this experiment condition.
