鱿鱼鱼精蛋白的提取工艺优化研究

摘要: 通过单因素和正交试验研究了鱿鱼鱼精蛋白的提取工艺条件,并对鱿鱼精巢的基本营养成分和鱿鱼鱼精蛋白进行了分析。结果表明,最佳工艺条件为硫酸浓度0.3 M、硫酸用量为3倍、提取次数2次、冷乙醇用量为3倍,在此条件下,鱿鱼鱼精蛋白的得率为3.42%,蛋白质含量达93%,且氨基酸组成丰富,碱性氨基酸占26.20%,其中精氨酸占9.61%,Tricine-SDS-PAGE电泳结果分析表明其组成成分比较复杂。

Abstract: Abstract:The single factor and orthogonal array design experiments of squid protamine extraction conditions ,the basic nutrients of squid testes and the squid protamine were studied. The results showed that the yield of squid protamine was 3.42% in the optimal condition of 3 times volume of 0.3M sulfuric acid concentration to extract protamine, repeating for 2 times, and 3 times volume of cold ethanol. The squid protamine contained 93% protein and was rich in all kinds of amino acids in which basic amino acids accounted for 26.2% and arginine accounted for 9.61%. According to the analysis of Tricine-SDS-PAGE electrophoresis, the molecular weight of squid protamine was less than 20 000Da.
