蜈蚣藻多糖纯化及抗氧化作用的研究

Study on purification and antioxidation of polysaccharides from Grateloupia filicina

摘要:

采用热水浸提法提取蜈蚣藻多糖,通过对超氧阴离子(?O2-)、羟基自由基(?OH)清除效果的研究评价了蜈蚣藻多糖的体外抗氧化活性,以及对蛋白脱除率和多糖损失率的分析比较了TCA法和Sevag法脱蛋白的效果,利用Sephadex G-150柱纯化了蜈蚣藻多糖,并分析了其部分理化性质和相对分子质量分布。结果表明,蜈蚣藻多糖对?OH清除率的IC50为4.62 mg/mL,对??O2-清除率的IC50为0.62 mg/mL,具有体外抗氧化效果;Sevag法脱蛋白效果优于TCA法,蛋白脱除率达85.55%,多糖损失率为15.93%;蜈蚣藻多糖是一种以多糖为主、含有糖蛋白的复合物,其相对分子量的分布范围在3.5×105~1.2×105。

Abstract:

By studying cleaning effect of ?O2- and ?OH to asssess vitro antioxidation and analysing deproteinization rate and polysaccharides loss rate to compare trichloroacetic acid (TCA) method with Sevag method of polysaccharides extracted by water extraction from grateloupia filicin.Using glucan gel G-150 chromatography column to purify polysaccharides and determine the distribution of molecular weight, furthermore analyzing some of its physical and chemical properties.The results indicate that polysaccharides from grateloupia filicin has antioxidant activity, IC50 on scavenging ?OH is 4.62 mg/mL,IC50 on scavenging ?O2- is 0.62 mg/mL.On effect of deproteinization , Sevag method is superior to TCA method, deproteinization rate is 85.55% and polysaccharides loss rate is 15.93%. Polysaccharides from grateloupia filicin is a kind of compounds with major constituent polysaccharides and glycoprotein,and the distribution of molecular weight is between 3.5×105 aand 1.2×105.
